Abstract:

Tato diplomová práce se soustřeďuje na optimalizaci a zavedení metodiky pro určení genotypu a následně i fenotypu barvy očí a vlasů u vybraných jedinců z velkomoravského pohřebiště Znojmo-Hradiště. Teoretická část navazuje na autorčinu bakalářskou práci, ve které jsou zpracovány poznatky o fenotypu zrzavých vlasů a polymorfismech genu MC1R podmiňujících tento fenotyp. Tyto polymorfismy jsou součástí HIrisPlex systému, jež se zdá být vhodným predikčním systémem pro fenotypování barvy vlasů a očí jak u recentního člověka, tak u historických pozůstatků. Cíle práce byly splněny v rámci praktické části, ve které se povedlo určit genotyp čtyř jedinců z deseti ze zmíněného pohřebiště pomocí analýzy sedmnácti jednonukleotidových polymorfismů HIrisPlex systému. U tří jedinců bylo možné z jejich genotypu predikovat, zda měli světlé nebo tmavé vlasy a také jakou měli nejpravděpodobnější barvu očí a vlasů. U jednoho jedince bylo možné predikovat pouze světlé či tmavé vlasy a barvu očí. Po prvotní optimalizaci metod na starobylou DNA byly genotypy určeny následnou analýzou prostřednictvím sekvenování nové generace. Fenotypy bylo možné predikovat s využitím interaktivní webové stránky založené na HIrisPlex systému. Výsledné fenotypy pak slouží pro dokreslení představy o vzhledu starých Slovanů.

Abstract:

This thesis is focused on the optimization and implementation of methods for determining the genotype and subsequently the eye and hair color phenotype in selected individuals from the Moravian burial site Znojmo-Hradiště. The theoretical part follows up on the author's bachelor's thesis, which is focused on the red hair phenotype and polymorphisms of the MC1R gene determining this phenotype. These polymorphisms are part of the HIrisPlex system, which could be a suitable system to predict hair and eye color phenotype in both recent humans as well as in historical remains. The goals of the work were met in the practical part of the thesis, which successfully determined the genotype of four out of ten individuals from the above-mentioned burial site by analyzing seventeen single nucleotide polymorphisms of the HIrisPlex system. From the genotype of these three individuals it was possible to predict, whether they had light or dark hair and their most likely eye and hair color. For one individual, only light or dark hair and their eye color could be predicted. After the initial ancient DNA methods optimization, the genotypes were determined using subsequent analysis through next generation sequencing. Phenotypes could be predicted using an interactive website based on the HIrisPlex system. The resulting phenotypes then serve to illustrate the possible appearance of the ancient Slavs.
