Mutual interactions between the cell cycle and ciliogenesis

Abstract:

Tato bakalářská práce se zabývá primárním ciliem a vztahem mezi buněčným cyklem a ciliogenezí. Primární cilium je nepohyblivá organela s mikrotubulární strukturou vyskytující se téměř u všech typů zdravých buněk. Ve většině případů vyčnívá z buněčného povrchu, avšak někdy vyčnívá pouze částečně nebo je kompletně uvnitř buňky. Primární cilium je senzorickým centrem buňky, které se podílí na regulaci proliferace a embryonálního vývoje. Skládá se z bazálního tělíska, axonemy a ciliární membrány. Obvykle je přítomno v G0/G1 fázi buněčného cyklu a po vstupu buňky do buněčného cyklu je resorbováno. Ve většině případů se ciliogeneze a postup v buněčném cyklu vzájemně vylučují a resoprce cilia je nezbytná pro buněčné dělení. Je známo několik proteinů hrajících roli v regulaci ciliogeneze i buněčného cyklu a narušení jejich exprese vede k poruchám tvorby a funkcí primárního cilia, poruchám buněčného cyklu nebo obojího současně což naznačuje, že tyto procesy jsou ve velmi úzkém vzájemném vztahu. Tyto proteiny řídí rozhodnutí, zda dojde k resoprci a návratu buňky do buněčného cyklu. Defekty primárního cilia mají širokou škálu patologických projevů od mírných až po fatální, souhrnně se tyto nemoci nazývají ciliopatie. Ještě pár desítek let zpět byla tato organela považována za nedůležitou, v posledních letech se však ukazuje, že má pro buňku velký význam.

Abstract:

This bachelor thesis is focused on a primary cilium and the relationship between the cell cycle and ciliogenesis. The primary cilium is a microtubule-based non-motile organelle that occurs in almost all types of normal healthy cells. The primary cilium usually emerges from the cell surface, but in some cases emerges only partially or is even un-emerged inside the cell. The primary cilium is the sensory center of the cell that participates in the regulation of the cell proliferation and embryonic development. The primary cilium is composed of a basal body, microtubule axoneme, and ciliary membrane. Usually is the primary cilium present in G0/G1 phase of the cell cycle and after re-entry of the cell cycle is resorbed. In most case, ciliogenesis and the cell cycle progression are mutually exclusive, and the resorption of the primary cilium is necessary for the cell division. Several proteins are known for their role in the regulation of both ciliogenesis and the cell cycle and impair of their expression usually leads to defects in the primary cilium or the cell cycle or both which suggest their close relationship. These proteins control decision whether the primary cilium will be resorbed and cell will re-enter the cell cycle. Disorders in the primary cilium manifest in a broad spectrum of pathologies, from the mild ones to the fatal once, and these manifestations are collectively called ciliopathies. Only a few decades ago, this organelle was considered as unimportant, but lately it has been shown that primary cilium is really important for cell.
