Genes of innate immunity and their effect on fitness in birds

Abstract:

V posledním desetiletí se geny zúčastněné v imunitní odpovědi staly předmětem zvýšeného zájmu v oblasti studia srovnávací a evoluční biologie, a to i u jiných živočišných taxonů, než modelových druhů obratlovců. Většina prací na toto téma se týká genů hlavního histokompatibilního komplexu (MHC), které představují zásadní složku získané imunity u obratlovců. O vrozené imunitě a jejích genech je na druhou stranu známo pouze velmi málo informací, zejména u volně žijících obratlovců. Stěžejním nespecifickým imunitním mechanizmem je rozpoznání evolučně konzervovaných struktur patogenů (mikroorganizmů a virů), tzv. PAMPs, pomocí receptorů buněk imunitního systému - PRRs, které následně umožňuje spuštění obranné imunitní odpovědi. K nejvýznamnějším a současně nejlépe prostudovaným receptorům vrozené imunity patří Toll-like receptory (TLRs), na něž je předkládaná práce z velké části zaměřena. Kromě TLRs existuje celá řada jiných receptorů vrozené imunity, tyto jsou ale podstatně méně prostudované. V současné době taktéž není k dispozici mnoho údajů o souvislosti vnitrodruhové variability TLRs jednotlivých složek fitness, a to ani u savců a ptáků, kteří jsou hlavními modelovými skupinami evolučně-ekologického výzkumu. Hlavním cílem této práce je (1) vyhledat všechny dostupné informace o struktuře a rozmanitosti TLRs a ostatních genů vrozené imunity ptáků a (2) zaměřit se na možnou souvislost variability genů vrozené imunity a zdatnosti (fitness) u ptáků.

Abstract:

In the last decade, genes important for proper immune response have become subject of great interest in the field of comparative and evolutionary biology, even outside the model vertebrates. Most of the immunogenetic studies deal with Major histocompatibility complex (MHC) genes, which represent a crucial component of the vertebrate acquired immune system. On the other hand, very little is known about the structure and variation of innate immunity genes in wild vertebrates. A key mechanism of innate immunity is the recognition of evolutionarily conserved structures of pathogens (microorganisms and viruses), called PAMPs, via receptor cells of the immune system - PRRs, that initiate immune response and defence of organism. The most important and the best-studied receptors of innate immunity are Toll-like receptors (TLRs), which are the main focus of this thesis. In addition to TLRs, there are a number of other receptors of innate immunity, but these are much less studied. So far, there are almost no data also about the linkage between the intraspecific variation of TLRs and fitness components, even for mammals and birds, which are among the most important models of evolutionary ecology research. The aims of this study are (1) bibliographic search for all available information on the structure and diversity of TLRs and other genes of innate immunity in birds and (2) looking for the possible linkage between their variation and fitness in birds.
