The ability of bifidobacteria to utilize artificial sweeteners and sugar alcohols as a sole energy source

Abstract:

Obsah této práce byl zaměřen na testování schopnosti bifidobakterií utilizovat umělá sladidla a cukerné alkoholy jako jediný zdroj energie. Předpokladem práce bylo, že schopnost metabolizovat tyto látky budou mít pouze některé druhy bifidobakterií a že tato vlastnost bude kmenově specifická. Obecně jsou umělá sladidla velmi kontroverzním tématem ve vědecké komunitě a společnosti. Stále není natolik průkazné, zda role umělých sladidel v lidské výživě má pozitivní či negativní vliv na střevní mikrobiotu a zdraví lidského organismu obecně. Praktická část práce probíhala v in vitro podmínkách, kdy bylo otestováno celkem 48 kmenů rodu Bifidobacterium pomocí makrodiluční a mikrodiluční metody. Jako substráty pro růst bifidobakterií sloužily 4 umělá sladidla (acesulfam K, aspartam, sacharin, sukralóza) a 4 cukerné alkoholy (erythritol, isomalt, sorbitol, xylitol), které se v koncentraci 1,5 g/l přidávaly do API 50 CHL média. Pro kontrolu nárůstu bifidobakterií byla použita i pozitivní kontrola se substrátem v podobě glukózy a negativní kontrola bez substrátu. Kultivace bifidobakterií probíhala při teplotě 37 °C po dobu 24 až 48 hodin. Vyhodnocení experimentu se lišilo podle použité metody. U obou metod se hodnotila optická denzita a pH. Měření optické denzity však u každé metody probíhalo odlišně. U makrodiluční metody byla vyhodnocena optická denzita pomocí McFarlandovy zákalové stupnice, u mikrodiluční metody byla vyhodnocena pomocí přístroje Tecan Infinite M200. Z výsledku bylo patrné, že u makrodiluční metody umělo utilizovat daný substrát 10 kmenů bifidobakterií a u mikrodiluční metody 12 kmenů bifidobakterií z celkového počtu 48 testovaných kmenů. Nejčastěji metabolizované byly cukerné alkoholy sorbitol a isomalt. Z umělých sladidel to byly acesulfam K a aspartam. Ostatní sladidla (erythritol, sacharin, sukralóza a xylitol) nebyly metabolizovány bifidobakteriemi, čímž se potvrdila část hypotézy, že testovaná sladidla metabolizovaly jen některé druhy bifidobakterií. Vlastnost bifidobakterií utilizovat různá umělá sladidla a cukerné alkoholy se potvrdila i u různých kmenů jednoho druhu bifidobakterií. Např. 3 kmeny u druhu B. catenulatum. Kmen B. catenulatum D1 a B. catenulatum D16 byly schopné utilizovat sorbitol v makrodiluční metodě. B. catenulatum D36 kromě sorbitolu dokázalo v této metodě utilizovat i isomalt. Z těchto výsledků lze vyvodit potvrzení zbylé části hypotézy, že schopnost metabolizovat testovaná sladidla je u bifidobakterií kmenově specifická. Z hlediska informací z odborné literatury shromážděných v této práci a z výsledků praktické části této práce nelze zcela s jistotou říct, zda schopnost metabolizovat umělá sladidla a cukerné alkoholy má pozitivní či negativní vliv na střevní mikrobiotu a lidské zdraví. Lze tedy tyto domněnky aplikovat v budoucnu při dalším výzkumu v této oblasti.

Abstract:

The aim of this thesis was focused on testing the ability of bifidobacteria to utilize artificial sweeteners and sugar alcohols as the only source of energy. The premise of the work was that only some species of bifidobacteria will have the ability to metabolize these substances and that this property will be strain specific. In general, artificial sweeteners are controversial topic in the scientific community and society. It is still not clear whether the role of artificial sweeteners in human nutrition has a positive or negative effect on the intestinal microbiota and the human health. The experiment of the thesis tooks place in vitro when a total of 48 strains of the genus Bifidobacterium were tested using macrodilution and microdilution methods. Four artificial sweeteners (acesulfame K, aspartame, saccharin, sucralose) and four sugar alcohols (erythritol, isomalt, sorbitol, xylitol) were used as substrates for the growth of bifidobacteria, which were added to API 50 CHL medium at concentration of 1.5 g / l. The positive control with substrate in the form of glucose and the negative control without substrate were also used to control the growth of bifidobacteria. The bifidobacteria were cultivated at 37 ° C for 24 to 48 hours. The evaluation of the experiment differed according to used method. Both methods were evaluated for optical density and pH. However, the measurement of optical density was different for each method. For the macrodilution method, the optical density was evaluated using the McFarland scale, for the microdilution method it was evaluated using the Tecan Infinite M200. The results showed that in the macrodilution method 10 bifidobacterial strains could utilize the given substrate and in the microdilution method 12 bifidobacterial strains out of the total number of 48 tested strains could utilize the substrates. The most metabolized sugar alcohols were sorbitol and isomalt, the most metabolized artificial sweeteners were acesulfame K and aspartame. Other sweeteners (erythritol, saccharin, sucralose, xylitol) were not metabolised by bifidobacteria, confirming part of the hypothesis that the tested sweeteners were metabolized only by some species of bifidobacteria. The ability of bifidobacteria to utilize different artificial sweeteners and sugar alcohols has also been confirmed in different strains of one species of bifidobacteria. E.g., 3 strains in B. catenulatum. B. catenulatum D1 and B. catenulatum D16 were able to utilize sorbitol in the macrodilution method. In addition to sorbitol, B. catenulatum D36 was able to utilize isomalt in this method. From these results, it is possible to confirm the rest of the hypothesis that the ability to metabolize the tested sweeteners is strain-specific in bifidobacteria. From the point of view of the information gathered in this thesis and from the results of our experiment, it is not possible to say with certainty whether the ability to metabolize artificial sweeteners and sugar alcohols has a positive or negative effect on the intestinal microbiota and human health. Therefore, these assumptions can be applied in future research.
