Molecular, biochemical and immunological characterization of cystatins of the parasitic Nematodes from the genus Trichinella

Abstract:

Hlístice rodu Trichinella jsou paraziti infikující trávicí a svalovou soustavu. Po celém světě napadají široké spektrum hostitelů, jako jsou ptáci, plazi a savci. Sedm z dvanácti známých genotypů u člověka způsobují závažné a často smrtelné onemocnění zvané trichinelóza. Svalové larvy zástupců rodu Trichinella při interakci s hostitelem vylučují do svého okolí exkrečně-sekreční produkty (E-S produkty). Jedná se o směs molekul, které se podílí na mnoha významných biologických procesech, jako je vývoj, růst, migrace, trávení a především ovlivňování hostitelské imunitní reakce mířené proti parazitovi. Mezi proteiny zastoupenými ve vysoké koncentraci v E-S produktech byly identifikovány mimo jiné i cystatiny. Cystatiny jsou reverzibilní inhibitory původně zacílené na cysteinové peptidázy skupiny C1 a C13. Sdružují velkou skupinu evolučně příbuzných enzymů, které sdílí určitou sekvenční a strukturální podobnost. Tato diplomová práce je zaměřena na “multi cystatin-like domain” (mcd-1) geny u druhů Trichinella spiralis, T. britovi a T. pseudospiralis. Byly získány kompletní sekvence mcd-1 genů všech tří modelových organizmů. Porovnáním dat byly určeny tři cystatinové domény u druhu T. spirals i T. britovi a dvě domény u druhu T. pseudospiralis. Všechny domény jsou si podobné a nesou specifické konzervované aminokyseliny, kterými jsou N-koncový glycin, čtyři cysteiny, P(S/T) motiv a SSTMG/GxxMG motiv. Rekombinantní analogy MCD-1 proteinů byly připraveny s využitím expresního systému Escherichia coli, což bylo potvrzeno metodou hmotnostní spektrometrie MALDI TOF/TOF. V závěru byla prokázána fragmentace MCD-1 proteinů na části o různém počtu domén.

Abstract:

The nematodes of the genus Trichinella are intracellular parasites which invade the digestive and muscular system. They infect a broad range of worldwide distributed hosts like birds, reptiles and mammals. All Trichinella spp. are zoonotic but only seven of twelve genotypes have been detected in humans, causing the trichinellosis - a serious, sometimes fatal disease. During the infection, muscle larvae of Trichinella spp. release excretory-secretory (E-S) products into the external enviroment. These molecules are involved in many essential biological processes like development, growth, migration, digestion and also in manipulation of the host immunne system which is targeted against the parasite. One of the dominant components of E-S products are cystatins. Cystatins are a reversible inhibitors of C1 and C13 cysteine peptidases. They comprise a large group of evolutionarily related proteins which share a specific sequential and structural similarity. This thesis is focused on characterisation of „multi cystatin-like domain“ (mcd-1) genes of three trichinella species - Trichinella spiralis, T. britovi and T. pseudospiralis. The complete sequences of mcd-1 genes were obtained. The comparison of sequential data showed that cystatin genes of T. spiralis and T. britovi are composed of three cystatin domains while T. pseudospiralis gene is formed by two domains. All domains are characterized by highly conserved amino acids which are N-terminal glycine, four cysteine, P(S/T) motiv and SSTMG/GxxMG motiv. Recombinant analog of MCD-1 proteins were prepared in Escherichia coli expression system. The presence of the desired protein was identified by the mass spectrometry method MALDI TOF/TOF. The fragmentation of MCD-1 proteins into the parts with different numbers of domains was confirmed.
