Genetic structure of annual Nothobranchius fishes from southern Mozambique

Abstract:

Rod Nothobranchius zahrnuje drobné, krátkověké ryby obývající izolované periodické tůně v savanách východní Afriky. Všechny druhy jsou anuální; dospělci umírají během období sucha a nová generace přežívá ve formě jiker uložených ve vyschnutém substrátu. Cílem této práce bylo analyzovat genetickou variabilitu a detailní prostorovou strukturu mezi populacemi v jižním a středním Mozambiku a provést detailní srovnávací analýzu pro testování alternativních hypotéz kolonizace této oblasti a disperze halančíků (šíření dospělců během rozsáhlých záplav versus disperze jiker na tělech velkých savců). Byla analyzována populační genetická struktura tří druhových komplexů (N. furzeri/kadleci, N. orthonotus komplex a N. rachovii komplex) z jižního okraje areálu r. Nothobranchius. Jejich obdobná distribuce a nároky na habitat je činí ideálním modelem pro komparativní studie. Byla použita sada 12-13 mikrosatelitových lokusů pro 1638 jedinců z 96 populací a sekvence mitochondriálního genu pro cytochromu b pro 463 jedinců ze 152 populací. Tyto vzorky pokrývají celý dosud známý areál rozšíření studovaných taxonů. Genetické rozdíly mezi jednotlivými populacemi byly většinou velmi výrazné, a to i v případě lokalit vzdálených pouze několik kilometrů. To naznačuje důležitou roli genetického driftu a nízkou schopnost disperze. Překvapivým zjištěním je role hlavních toků velkých řek (např. Save, Limpopo) jako bariér toku genů, což je u ryb vzácná situace srovnatelná se situací známou u suchozemských zvířat (populace ryb bývají strukturovány podle povodí, bariéry jsou typicky mezi povodími). Vysoká divergence i mezi některými sousedními lokalitami (konzistentně pro všechny tři komplexy) nasvědčuje tomu, že jikry nejsou transportovány na tělech migrujících býložravců a tok genů mezi lokalitami v rámci stejné pánve je zajišťován během občasných záplav. Výsledky srovnávací fylogeografie také identifikovaly hlavní refugia, ve kterých populace těchto savanových druhů přežívaly období vlhkého klimatu v průběhu Plio-Pleistocénu.

Abstract:

The genus Nothobranchius is a group of small fishes that inhabit temporary savannah pools in Eastern Africa. All species are annual; the adults die when the habitat desiccates and new generation survives in the form of eggs encased in the dry mud. The objective of this study was to analyse the genetic variability and detailed spatial structure among killifish's populations in Southern and Central Mozambique and to accomplish detailed comparative analysis for testing of alternative hypotheses about colonization of this area and Nothobranchius dispersal (adult dispersal during large floods versus egg dispersal in mud on bodies of large mammals). This work is focussed on population genetic structure of three species complex (N. furzeri/kadleci, N. orthonotus complex a N. rachovii complex) from the southern periphery of Nothobranchius range, using samples covering their entire known distribution area. Similar distribution and habitat requirements of the three complexes make them an ideal model for comparative studies. Set of 12-13 microsatellite loci genotyped in 1638 individuals from 96 populations and cytochrome b sequences from 463 fish representing 152 populations were used as genetic markers. Profound genetic differentiation among populations was very typical; in some cases even adjacent populations distant by few kilometres were genetically very distinct, suggesting a significant role of genetic drift and low ability of dispersion. Surprisingly, large rivers (e.g. Save, Limpopo) formed major barriers to gene flow. This situation has been rarely described for fish taxa and it is comparable to that known for terrestrial animals (usually fish populations tend to be structured according to the river basins, barriers to gene flow occur among rivers). High divergence even between neighbouring populations (consistent for several species) suggests that the eggs are not transported by large mammals and the gene flow is likely mediated by occasional flooding within one basin. The results of comparative phylogeography identified the main refugia of these savanna fishes, in which the populations survived periods of humid climate during the Plio-Pleistocene.
