Experimental application of biochemical markers in model vertebrates

Abstract:

Biochemické markery mohou sloužit jako důležité a citlivé ukazatele účinků stresorů na organismus. Práce je zaměřena na stanovení biomarkerů oxidativního stresu, který jakožto obecný mechanismus umožňuje posuzovat vlivy různých stresorů a expozic. Často zpracovávaným tématem je problematika vodního květu a s ním související expozice sinicemi, zvláště pak jejich toxiny. V první teoretické části práce je zpracována rešerše účinků sinicové biomasy na křepelku japonskou (Coturnix japonica). V praktické části je pak provedeno stanovení biomarkerů glutathionu, glutathion-S-transferázy a lipidní peroxidace na tomto modelovém organismu po chronické expozici sinicovému vodnímu květu. Druhá část práce je zaměřena na inhalační expozici nanočásticemi. Nanočástice jsou již dnes široce používány pro různé účely, ale o jejich biologických účincích není dosud známo dostatek informací. V rešeršní části práce jsou zpracovány podklady z literatury zabývající se inhalační expozicí a jejím vlivem na oxidativní stres u organismů, v praktické části je provedeno a vyhodnoceno stanovení biomarkerů glutathionu, glutathion-S-transferázy a lipidní peroxidace na myši obecné (Mus musculus) exponované inhalačně nanočásticemi manganu. V praktické části práce byla zahrnuta charakterizace vybraných metod a pomocí absorpčních spekter ověřena čistota produktů. Bylo zjištěno ovlivnění parametrů spojených s klíčovým neenzymatickým antioxidantem glutathionem po působení sinicové biomasy s obsahem microcystinů u modelových ptáků. Tyto parametry byly ovlivněny také při dlouhodobější inhalaci nanočástic u savců, která navíc vedla k oxidativnímu poškození lipidů v játrech i v mozku. Výsledky potvrzují univerzálnost oxidativního stresu jako důležitého toxického mechanismu pro různé typy stresorů, neboť u obou studovaných velmi rozdílných typů expozic byly zjištěny projevy působení oxidativního stresu.

Abstract:

Biochemical markers can serve as important and sensitive signals, that can indicate effects of various stressors on organism. This work is focused on the determination of biomarkers of oxidative stress, which is generous mechanism, that can be involved ineffects of various stressors and exposures. Frequently discussed theme is water bloom and connected exposure to cyanobacteria and their toxins. First part of this work contains review concerned with effects of cyanbacterial biomass on Japanese quail (Coturnix japonica). Analysis of biomarkers glutathione, glutathione-S-transferase and lipid peroxidation in this model organism chronically exposed to cyanobacterial biomass are realized in practical part. Second part is focused on inhalation exposure to nanoparticles. Nanoparticles are widely used for different applications but there is not enough information about their biological impacts. This thesis summerases literature about inhalation exposure to nanoparticles and its effects on oxidative stress in organisms. Analysis of biomarkers glutathione, glutathione-S-transferase and lipid peroxidation in mice (Mus musculus) exposed via inhalation of manganese nanoparticles are realized in practical part. The practical part included also the characterization of selected methods and purity of products was verified by measuring of absorption spectra. The results document effects on parameters related to the key nonenzymatic antioxidant glutatione after exposure of birds to cyanobacterial biomass. The same parameters were affected along with induction of lipid peroxidation in liver and brain after longer-term inhalation exposure to nanoparticles in mammals. These studies confirm the universal character of oxidative stress as an important toxic mechanism of different types of stressors, since for both very different types of exposure the results indicated involvement of oxidative stress.
