Donor’s age determination using Pyrosequencing methodology

Abstract:

Určení věku jedince spadá tradičně mezi úkoly antropologie. Ve druhé polovině 20. století se začaly objevovat další metody, založeny na biochemických, genetických a od konce 20. století také epigenetických procesech. Právě o nich – konkrétně o metylaci DNA, pojednává tato práce. V jejím rámci je testována metoda určení věku pro účely forenzní genetiky. Na základě úrovně metylace vytipovaných markerů lze z odebraného vzorku biologického materiálu donora odhadnout jeho věk. Tato informace je přínosná zejména v případě násilných trestných činů s neznámým pachatelem, kdy je každá informace o něm nedocenitelná a v mnoha případech může pomoci urychlit vyšetřování či zúžit okruh podezřelých. Metoda testovaná v této práci byla verifikována na krevních stopách. Na místě činu se však nachází také jiné biologické stopy, např.: sliny či sperma. Je proto snaha vyvíjet metodu pro určení věku na základě metylace DNA také pro jiné biologické materiály. Cílem této práce je ověřit, zda lze vyvinutou metodu aplikovat také na další biologické materiály. Úroveň metylace je specifická pro různé typy tkání a metody na ní založené nemusí fungovat univerzálně. Nicméně výsledky diplomové práce potvrdily, že při aplikaci metody na vzorky slin metoda funguje se srovnatelnou přesností. Oproti tomu, pro vzorky sper-matu metoda vhodná není. Je tomu tak právě proto, že se sperma od krve, na které byla metoda validována, co do složení velmi liší. Kromě vlivu zvoleného biologického materiálu je potřeba vzít v potaz také další faktory, které ovlivňují metylaci, a tudíž hodnotu výsledného odhadu věku. Jsou jimi například geografický původ, vysoký věk, zdravotní stav či životní styl jedince.

Abstract:

Estimation of human age had traditionally belonged among tasks of anthropology. In the second half of the 20th century, other methods, based on biochemical, genetic (and from the end of the 20th century), and epigenetic processes emerged. This thesis discusses the epigenetic approach, namely methylation of DNA. In the frame of this thesis, a method for forensic age estimation is tested. Based on the methylation levels of selected markers, it is possible to analyse biological material and estimate the donor’s age. In the case of violent criminal acts where the offender is unknown is the information of their age priceless. In many cases, it can help with speeding up of investigation or narrowing the suspect’s pool. The method tested in this thesis has been verified on blood samples. However, there are other biological traces at the crime scene, for example, saliva or semen. That is why there are attempts to develop a method for forensic age estimation based on DNA methylation also for other biological materials. The aim of this study is to verify if it is possible to apply the earlier developed method to other biological materials. Levels of methylation are tissue-specific, so DNA methylation-based method does not have to work universally. However, conclusions of this diploma thesis confirmed that when applied to saliva method work as accurately as on blood samples. By contrast, this method is not suitable for semen samples. It is because semen and blood, on which the method was validated, have diverse cell compositions. Apart from the impact of biological material, there are more factors, that can affect methylation and thus final estimated age, and must be taken into consideration. Such as geographical origin, high age, state of health, or lifestyle of an individual.
