Lectins as a tool for specific detection and glycoprotein purification

Abstract:

Tato práce byla zaměřena na problematiku přípravy afinitních sorbentů pro specifickou izolaci glykoproteinů. Pro vytvoření afinitních sorbentů, které vykazují specifické schopnosti dělení, tzn. že jsou schopny separovat rozdílné glykoproteiny, bylo využito imobilizace lektinů na komerční sorbent Epoxy- Sepharosu. Použité lektiny se vyznačovaly rozdílnými afinitními vlastnostmi k sacharidům. Očekávalo se tedy, že jejich spektrum separace bude odlišné. Tato práce se zabývá nejprve kultivací buněk E.coli nesoucích plastidy požadovaných proteinů. Následně expresí lektinů, jejich purifikací a také charakterizací vazebných a afinitních vlastností pomocí izotermální titrační mikrokalorimetrie (ITC) a rezonance povrchových plasmonů (SPR). Po izolaci 13 mg lektinu BclA a 20 mg lektinu RSL a po ověření jejich vazebných vlastností, byly lektiny podrobeny imobilizaci na komerční aktivovaný sorbent Epoxy- Sepharosu. Imobilizace probíhala převážně za podmínek určených výrobcem. Připravené sorbenty, fungující na principu afinitní chromatografie, byly použity k separaci glykoproteinů ze dvou různých zdrojů. Pro základní testování vazebných vlastností afinitních kolon bylo použito komerční lidské sérum LYO HUM a následně byl využit také cytosol kvasinky Pichia pastoris. Výsledky testování nebyly pro námi zvolené použité lektiny BclA (z Burkholderia cenocepacia) a RSL (z Ralstonia solanacearum) úspěšné. Bylo proto využito dvou kontrolních lektinů, vykazující podobné vlastnosti s lektiny RSL a BclA, a to lektinu PA-IIL z Pseudomonas aerigunosa a AFL z Aspergillus fumigatus. Byla také optimalizována samotná metoda separace glykoproteinů. Nakonec se podařilo získat metodu, která ukázala pozitivní výsledky, zejména pro kontrolní lektin PA-IIL. Podařilo se tedy separovat glykoproteiny z krevního séra LYO HUM. Pozitivnější výsledky následně přinesla několikadenní regenerace sorbentu s imobilizovanými Ca2+ -dependentními lektiny BclA a PA-IIL. Pro méně známý lektin AFL byly všechny získané výsledky negativní, jelikož tento lektin nebyl schopen kovalentí vazby se sorbentem. Nejúspěšnější výsledky pro připravené afinitní sorbenty přinesla metoda, která využila supernatantu z kvasinky Pichia pastoris. U této metody se projevily pozitivně všechny separace pro použité lektiny RSL, BclA a také pro kontrolní lektin PA-IIL.

Abstract:

The diploma thesis focuses on preparation of affinity columns for specific isolation of glycoproteins. To produce affinity columns that show specificity - this means that they are able to separate different glycoproteins – the immobilisation of lectins on commercial resin Epoxy – Sepharose was used. The carbohydrate-binding proteins (lectins) that were used displayed different specificity and affinity for carbohydrates. It was expected that their abilities to separate glycoproteins will be different. First of all, this thesis deals with the cultivation of E. coli cells harbouring plasmids coding used proteins. Consequently expression of lectins, their purification and characterization of binding affinity properties by using isothermal titration microcalorimetry and surface plasmon resonance was performed. After the isolation of 13 mg lectin BclA (from Burkholderia cenocepacia) and 20 mg lectin RSL (from Ralstonia solanacearum) and after the verification of their binding properties, the lectins were immobilized on a commercial activated resin Epoxy - Sepharose. The immobilization proceeded largely under the conditions specified by the producer. The prepared columns based on the principle of affinity chromatography were used for preparation of glycoproteins from two different sources. The functionality of prepared affinity columns was tested using the control human blood serum LYO HUM and subsequently, the cytoplasmic fraction of Pichia pastoris yeast was used. The results of testing the columns with immobilized lectin BclA and lectin RSL were not successful. Therefore, the two other control lectins were used. It was the lectin PA-IIL from Pseudomonas aeruginosa and the lectin AFL from Aspergillus fumigatus. They have similar properties as lectins BclA and RSL. Glycoprotein separation was further optimised. Finally, it was managed to get the method, which showed positive results, especially for the control lectin PA-IIL. The separation of glycoproteins from the blood serum LYO HUM was successful. Even more successful results were obtained using the several-day regeneration of columns with immobilized Ca2+ - dependent lectins BclA and PA-IIL. As for the less-investigated lectin AFL, all the obtained results were negative since this lectin was not bound on resin covalently. The best results were seen using cytoplasmic fraction of Pichia pastoris as a sample as all three lectins BclA, RSL and PA-IIL were able to separate glycoproteins.
