Cloning and protein expression of polyvalent staphylococcal bacteriophage encoded structural genes

Abstract:

Bakteriofág 812 je polyvalentní striktně virulentní fág, který je řazen do čeledi Myoviridae. Fág 812 a jeho mutanty vykazují silnou lytickou aktivitu proti širokému spektru kmenů Staphylococcus aureus a řadě dalším stafylokokovým druhům. Cílem diplomové práce bylo klonování a exprese čtyř strukturních proteinů bakteriofága 812 K1-420 kódovaných geny ORF72, ORF127, tmpH a ORF85. Na základě analýzy hmotnostní spektrometrií jsou tyto proteiny pravděpodobně součástí bičíku a bazální destičky fága, nicméně jejich přesná funkce je zatím neznámá. Za účelem purifikace pomocí metaloafinitní chromatografie byly proteiny při přípravě modifikovány tak, aby nesly histidinovou kotvu. Exprese rekombinantních proteinů probíhala v buňkách E. coli BL21 (DE3). Po přečištění proteinů metodou metaloafinitní chromatografie následovala purifikace pomocí gelové chromatografie, která zajistila dostatečnou čistotu gp72 a gp127 pro strukturní analýzu prostřednictvím krystalografie. Pro přípravu proteinů TmpH a gp85 byl připraven funkční genový konstrukt a proveden skríning podmínek exprese. Určení trojrozměrné struktury prostřednictvím krystalografie by mohlo významně přispět k detailnějšímu popisu stavby fága a pochopení funkcí samotných proteinů.

Abstract:

Polyvalent staphylococcal bacteriophage 812 is a stictly virulent phage belonging to the family Myoviridae. Phage 812 and its mutants are capable to lyse a broad host range of Staphylococcus aureus strains and act against another staphylococcus species. The aim of the diploma thesis was cloning and expression of four bacteriophage 812 K1- 420 structural proteins encoded by genes ORF72, ORF127, tmpH and ORF85. These four proteins are probably a structural components of the phage tail tube and baseplate as evidenced by MS analysis. Nonetheless, the precise function of these proteins is still unknown. For the purpose of purification by metallo-affinity chromatography the proteins have been modified to carry a polyhistidine tag. The recombinant proteins were expressed in E. coli BL21 (DE3) cells and purified using metallo-affinity chromatography. For acquisition a higher purity of proteins the next step was purification by gel filtration. The purified proteins were used for determination of their three-dimensional structure using crystallography methods, which might contribute to better description of the phage structure and the understanding of the functions of these proteins.
