The role of Dishevelled in the G2/M phase of the cell cycle

Abstract:

Signalizace Wnt je jednou z několika signálních drah, které jsou nezbytné pro správný embryonální vývoj a pro udržení homeostázy během života jedince. Ligandy Wnt po navázání k receptoru aktivují signalizační kaskádu například až k transkripci cílových genů. Jednou z hlavních komponent přenosu signálu od receptorů aktivovaných prostřednictvím ligandů Wnt k cílovým efektorům je protein Dishevelled, jehož funkce je regulována posttranslačními úpravami, ale i interakcí s různými vazebnými partnery. V posledních letech se ukazuje, že se zřejmě nepodílí pouze na přenosu signálu pro signalizaci Wnt, ale že by mohl být zapojen i do jiných dějů, které se signalizací Wnt nesouvisejí. Cílem této diplomové práce je identifikace a ověření vazby Dishevelled a kandidátů ze skupiny převážně centrozomálních proteinů, dále také zkoumání vlivu Dishevelled na průběh buněčného cyklu. Zjistili jsme, že se Dishevelled váže k některým proteinům specifickým pouze pro centrozom, významně ovlivňuje průběh buněčného cyklu a mohl by tak být zapojen do řízení buněčného dělení.

Abstract:

Wnt signalling is one of the few signalling pathways which are necessary for the proper embryonic development and for maintenance of homeostasis during adult life. After binding to receptor, Wnt ligands activate signalling cascades with effect on e.g. gene transcription. Dishevelled is one of the key components in the transduction of signal from ligand activated receptors and its function is regulated by posttranslation modifications or interaction with various binding partners. The investigations in few last years showed that Dishevelled is not solely confined to signal transduction during the Wnt signalling but can also be involved in processes unrelated to this signalling pathway. The aim of this thesis is the identification and confirmation of binding of previously unrelated candidates from the group of centrosomal proteins to Dishevelled and its possible impact on cell cycle regulation. We found out that Dishevelled binds to several centrosomal components that can have significant influence on cell cycle regulation and cell division itself.
