The role of uniparental disomy in human diseases

Abstract:

Tato práce je zaměřena na úlohu uniparentální dizomie při vrozených onemocněních člověka. První část práce se věnuje definici pojmu uniparentální dizomie, jejím typům a genomovému imprintingu, jež reguluje genovou expresi na základě modifikací DNA. Ve druhé části práce jsou popisovány mechanismy vzniku uniparentální dizomie od okamžiku fertilizace do ustanovení dizomického stavu chromozomů v zygotě. Třetí část představuje různé metody detekce uniparentální dizomie založené na srovnávání alel postiženého jedince a jeho rodičů, na určení methylačního stavu DNA a na analýze jednonukleotidových polymorfizmů. Hlavní část práce se soustředí na vrozená genetická onemocnění, jež vznikla mechanismy vedoucími k uniparentální dizomii. Jsou zmíněny genetické aspekty onemocnění a jejich fenotypový dopad.

Abstract:

This thesis is focused on the role of the uniparental disomy in human congenital diseases. The first part of thesis is concentrated on the definition of concept of uniparental disomy, its forms and genomic imprinting, that regulates a gene expression by modifications of DNA. In the second part, there are described mechanisms of formation of the uniparental disomy from the moment of fertilization to disomic constitution of chromosomes in zygote. In the third part there are shown different methods for detection of uniparental disomy. They are based on comparison of alleles of affected individuals and their parents, on determination of DNA methylation status and on analysis of single nucleotide polymorphisms. The main body of this thesis is concentrated on congenital genetic diseases, which were created by mechanisms leading to uniparental disomy. There are mentioned genetic aspects of diseases and their phenotype impacts.
