Abstract:

Rostoucí výskyt bakterií rezistentních k antibiotikům je závažným problémem současné medicíny. Nemožnost léčení některých infekcí antibiotiky vede k hledání alternativních způsobů léčby. Zde je snaha zavést fágovou terapii, která se ukazuje být efektivní při léčení bakteriálních infekcí. Dříve se užívalo celých fágových částic, dnes je zájem zaměřen spíše na fágem kódované antimikrobiální látky, jakými jsou například lytické enzymy. Tato práce byla zaměřena na lytické enzymy polyvalentního fága 812 a jeho mutanta 812F1 parazitujících na širokém rozmezí kmenů nebezpečného patogena jakým je Staphylococcus aureus. V diplomové práci byly definovány podmínky pro klonování a expresi genu pro endolyzin. Byla ověřována funkčnost klonovaných genů a produkty těchto genů byly blíže charakterizovány. Při ověřování vlastností endolyzinu se ukazuje, že rozmezí hostitelů lyzovaných fágovou částicí je výrazně menší než při použití endolyzinu. Potvrzuje se, že rezistence bakterií k fágu je způsobována jinými mechanismy než neschopností endolyzinu lyzovat bakteriální buňku, a tedy že rezistence k lytickým enzymům je velmi nepravděpodobnou příčinou rezistence buněk k fágům. Zkoumané endolyziny byly dále analyzovány pomocí SDS-PAGE, zymogramu nebo westernového přenosu a byly připravovány pro plánovanou purifikaci metalochelatační chromatografií. Bude třeba optimalizovat metody pro získání čistého enzymu. Endolyziny mají velký potenciál a mohly by být využity při fágové terapii pro léčení infekcí vyvolaných nejen bakteriálními kmeny rezistentními k antibiotikům.

Abstract:

Antibiotic resistant bacteria are significant problem in current medicine. Antibiotic treatment of infections of some pathogenic bacteria is inefficient in many cases so an alternative ways of therapy are trying to be found. Phage therapy is proven over the years as highly effective method to fight antibiotic resistant pathogenic bacteria. There were entire phage particles used in the past, but currently scientists are mostly focused on the antibacterial substances produced by bacteriophages, mostly on lytic enzymes. This diploma thesis is focused on lytic enzymes of polyvalent phage 812 and its mutant 812F1 which are able to infect a broad range of pathogenic Staphylococcus aureus strains. There were defined conditons for cloning genes for endolysin and solved some problems that occured during this procedure. Function of cloned genes were confirmed and protein products were further characterized. It turns out that endolysin has much wider host range then complete phage particle. It indicates that bacterial resistance to phage is caused by completely different mechanisms than by inability of endolysin to lyse bacteria. It is very unlikelythat the resistance to endolysins will occur. Endolysins were then characterized by methods like SDS-PAGE, zymogram or western blot. It was planed to purify enzymes by metal chelate affinity chromatography, but a lot of optimalization has to be done before a pure enzyme will be obtained. Endolysins have nevertheless a great potential and they can be effectively used in treatment of infections of patogenic bacteria not only resistant to antibiotics.
