Centrifugation Effect on Fish Blood Parameters

Abstract:

Bakalářská práce se zabývá rybí hematologií, a to konkrétně červeným krevním obrazem, u kterého byl zkoumán vliv centrifugace na hodnoty hematokritu a hemoglobinu. Práce zahrnuje literární rešerši na zkoumané druhy ryb, červený krevní obraz, odběry krve u ryb a centrifugaci. Praktická část se zabývá stanovením červeného krevního obrazu u zkoumaných druhů ryb. Mezi zkoumané druhy ryb patří kapr obecný (Cyprinus carpio), pstruh duhový (Oncorhynchus mykiss), keříčkovec červenolemý (Clarias gariepinus) a tlamoun nilský (Oreochromis niloticus). Od každého druhu byla 10 jedincům odebraná krev, ze které se následně stanovovaly hodnoty celkového počtu erytrocytů (RBC), koncentrace hemoglobinu (Hb), hematokritová hodnoty (PCV) a hodnoty výpočtové: střední objem erytrocytu (MCV), hemoglobin erytrocytu (MCH) a střední barevná koncentrace (MCHC). Byl sledován vliv vícenásobné centrifugace u hodnot hematokritu a u hemoglobinu byly porovnávány dvě hodnoty: vzorek centrifugovaný se vzorkem bez centrifugace. Nad rámec cílů práce byly sledovány hodnoty hemoglobinu v časových úsecích od 0. hodiny do 1 008. hodiny od odběru krve. Na těchto hodnotách jsme ověřovali vliv doby od odběru na hodnoty hemoglobinu. Data byla vyhodnocena u vlivu centrifugace na hematokritovou hodnotu a na koncentraci hemoglobinu pomocí dvouvýběrového T-testu a následně byly sestaveny krabicové grafy pro lepší zobrazení dat. U vlivu doby od odběru krve na hodnoty hemoglobinu bylo využito Wilcoxonova párového testu. Hodnoty z dlouhodobého měření byly vyneseny do krabicového grafu. Červený krevní obraz u pozorovaných jedinců vyšel z větší části v rozmezí standardních hematologických hodnot pro dané druhy ryb. Centrifugace statisticky významně neovlivnila ani hodnoty hematokritu, ani koncentrace hemoglobinu u sledovaných druhů ryb. U koncentrace hemoglobinu v závislosti na době odběru byla prokázána změna výsledku již po 24. hodině u všech pozorovaných druhů, s výjimkou pstruha duhového, kde byla pozorována změna až na 72. hodině. Opakovanou centrifugaci lze využít u keříčkovce červenolemého a tlamouna nilského u vzorků, u kterých dochází k nerovnoměrnému oddělení červených krvinek od plazmy. Při měření koncentrace hemoglobinu u ryb může by centrifugace vynechána, díky tomu může dojít k urychlení dané metody. Na základě dlouhodobého pozorování změn koncentrace hemoglobinu od odběru krve je doporučeno provádět měření co nejdříve, resp. do 24 hodin po odběru.

Abstract:

This thesis deals with fish hematology, it specialises in red blood count, focusing on hematocrit and hemoglobin values. This study includes information about the analysed fish species, red blood count, fish blood sampling and centrifugation. Research part involves determination of the red blood count in the examined fish species. These species include eurasian carp (Cyprinus carpio), rainbow trout (Oncorhynchus mykiss), african sharptooth catfish (Clarias gariepinus) and nile tilapia (Oreochromis niloticus). Blood samples were collected from 10 individuals from each species. These samples were then used to determine values of red blood cell count (RBC), hemoglobin concentration (Hb), packed cell volume (PCV) and derived erythrocyte parameters: mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC). The study observed the effect of multiple centrifugations on packed cell volume and two methods of sample preparation for hemoglobin measurement: half of the samples were centrifuged, and the rest were not. This study also contains hemoglobin values measured in different time periods from 0 to 1008 hours since blood collection. These values were used to examine the effect of extended collection time on hemoglobin concentration. The impact of centrifugation on hematocrit and hemoglobin concentration in collected data were evaluated by a two-sample t-test. Box plots were constructed for visual interpretation of the results. The Wilcoxon matched pairs test was used to determine the effect of extended collection time of the blood samples on the hemoglobin concentration values. The values from the long term measurement have been plotted in a box plot. The red blood counts of each sample were largely within the range of standard hematological values for the species. Centrifugation had no statistically significant effect on either hematocrit values or hemoglobin concentrations in studied fish species. For the hemoglobin concentration, a change in the result depending on the time of sampling was already observed after 24 hours in all observed species, except for rainbow trout, where a change was not observed until 72 hours. Repeated centrifugation may be used for african sharptooth catfish and nile tilapia for samples with uneven separation of red blood cells from plasma. Centrifugation of the blood sample can be omitted to speed up the method when measuring hemoglobin concentration in fish. Based on long term observation of changes in hemoglobin concentration since blood collection, it is recommended that measurements are taken as soon as possible or within 24 hours after blood collection.
