Testicular steroidogenesis and the impact of environemental contaminants

Abstract:

Látky vyskytující se v životním prostředí hrají pravděpodobně důležitou roli v celosvětovém snížení reprodukčního potenciálu u samců. Tato práce se věnuje problematice samčí neplodnosti se zaměřením na testikulární steroidogenezi a její regulaci u hlodavců a lidí během fetálního vývoje a v dospělosti. Zabývá se také mechanismy působení a účinky známých environmentálních kontaminantů na samčí reprodukční zdraví. Experimentální část této práce se zabývá bazální a indukovanou produkcí testosteronu Leydigovými TM3 buňkami v závislosti na složení média, koncentraci nasazených buněk a stáří buněčné linie v kultuře. Jako známé účinné induktory testikulární steroidogeneze na produkci testosteronu v Leydigových TM3 buňkách byly vybrány luteinizační hormon, lidský choriogonadotropin a forskolin. Lidský choriogonadotropin a forskolin produkci T nestimulovaly. Luteinizační hormon byl schopen indukovat produkci T, ovšem produkce nebyla zopakovaná v dalších experimentech. Ukázal se možný vliv stáří buněčné linie v kultuře. V následujících experimentech se bude třeba zaměřit na závislost vlastností buněčné linie TM3 v kultuře (fenotyp) a na její schopnosti produkovat bazální a indukovaný testosteron.

Abstract:

Compounds which are occurred in the environment are likely to play an important role in the worldwide reduction of male reproductive potential. This thesis deals with the problem of male infertility focusing on testicular steroidogenesis and its regulation in rodents and humans during fetal development and in adulthood. We also discuss the effects of known endocrine disruptors and their mechanisms of action on male reproductive health. The experimental part of this thesis deals with the basal and induced production of testosterone by the Leydig TM3 cells depending on the composition of the medium, concentration and the age of the cell lines in culture. As known and effective inductors of testicular steroidogenesis and production testosterone in Leydig TM3 cells were selected luteinizing hormone, human chorionic gonadotropin and forskolin. Human chorionic gonadotropin and forskolin did not stimulate the production of testosterone. Luteinizing hormone was able to induce the production of testosterone, but the production was not repeated in subsequent experiments. He pointed to the possible influence of the age of cell lines in culture. The following experiments will be focused on the dependency of the properties of cell lines in culture TM3 (phenotype) and their ability to produce basal and induced testosterone.
