Dental mesenchymal stem cells in continuously-growing mouse incisor – functional study.

Abstract:

Charakteristickou vlastností myších řezáků je jejich schopnost neustálého růstu po celý život jedince. Tento růst je ovlivňován aktivitou a dynamickými interakcemi mezi epiteliálními a mezenchymálními kmenovými buňkami, které jsou lokalizovány v apikální části zubu. V této oblasti populace dentálních mezenchymálních kmenových buněk produkují buňky dřeně a zejména dentin tvořící odontoblasty. Již dříve bylo popsáno, že tyto mezenchymální kmenové buňky nejsou zcela homogenní, nicméně teprve nedávno byly pomocí analýzy transkriptomu odhaleny mikropopulace dentálních mezenchymálních kmenových buněk, jejichž vlastnosti doposud nebyly popsány. Jednou z těchto mikropopulací se zabývá tato diplomová práce. S využitím metod mapování vývojových linií a imunohistochemie, byly v rámci této práce pozorovány poziční vlastnosti a klonální distribuce těchto kmenových buněk v jejich niche a také buněk, které se z nich diferencovaly. Na tyto experimenty dále navázala pozorování funkčních vlastností těchto buněk. Popsána byla jejich schopnost udržet inkorporovaný analog thymidinu v čase a pozorován byl také vliv cílené genetické ablace na jejich funkci.

Abstract:

A characteristic trait of mouse incisors is their ability to grow continuously throughout the life of the individual. This growth is influenced by the activity and dynamic interactions between epithelial and mesenchymal stem cells, which are localized in the apical part of the tooth. In this region, the dental mesenchymal stem cell population produces the cells of the pulp and, in particular, the dentin-forming odontoblasts. It has been previously described that these mesenchymal stem cells are not completely homogeneous, but only recently, transcriptome analysis has revealed micropopulations of dental mesenchymal stem cells whose properties have not been described so far. This thesis focuses on one of these micropopulations. Using lineage tracing and immunohistochemistry, the positional properties, and clonal distributions of these stem cells in their niche, as well as the cells that differentiated there, were observed. These experiments were followed by observations of the functional properties of these cells. Their ability to maintain the incorporated thymidine analogue over time was described and the effect of targeted genetic ablation was observed on their function.
