Lokalizace nových genů rezistence k Blumeria graminis f.sp. hordei u Hordeum vulgare pomocí DNA markerů

Localisation of resistance genes to Blumeria graminis f. sp. hordei in Hordeum vulgare ssp. spontaneum using molecular markers

Abstract:

Cílem práce bylo studium genetické determinace a lokalizace nových genů rezistence u nově identifikovaného zdroje odolnosti divokého ječmene (Hordeum vulgare ssp. spontaneum) k padlí travnímu, podmíněnému houbovým patogenem Blumeria graminis f. sp. hordei. Genetické mapování genů rezistence bylo realizováno pomocí SSR a CAPS markerů v generaci F2 vzniklé po křížení mezi ozimou odrůdou kulturního ječmene (Hordeum vulgare) Tiffany a zdrojem rezistence H. vulgare ssp. spontaneum PI466461, odolným ke 22 evropským patotypům Bgh s nejširším spektrem virulence. Z výsledků fytopatologických studií byla předpovězena přítomnost dvou genů rezistence a typ dědičnosti těchto genů byl určen jako dominantní a neúplně dominantní. Výsledky testu na alelizmus prokázaly, že jeden z genů se nachází v lokusu Mla na chromozomu 1HS. Lokalizace druhého genu byla vyhledána pomocí modifikované bulkové analýzy bez smísení DNA jednotlivých rostlin. Při této analýze bylo testováno 34 SSR markerů a CAPS marker RGH1aI1a ležící v lokusu Mla. Vazba s genem rezistence byla zachycena u těchto pěti markerů: RGH1aI1a, Bmac0213 a EBmac0501 na chromozomu 1H, Bmag0500 na chromozomu 6H a Bmag0021 na chromozomu 7H. U markeru Bmac0213 na chromozomu 1H nebyla testem ?2 potvrzena shoda se štěpným poměrem 1:2:1 pro kodominantní typ dědičnosti a tento marker byl vyřazen. Markery z chromozomových oblastí s předběžně zachycenou vazbou byly dále testovány na 97 rostlinách generace F2 s cílem určení mapové pozice hledaných genů rezistence. Výsledky molekulární analýzy jednotlivých rostlin generace F2 pro markery RGH1aI1a, EBmac0501, Bmac0032, Bmac0063 a Bmac0090 na chromozomu 1H, Bmag0500 a Bmac0316 na chromozomu 6H a Bmag0021, EBmag0794, Bmag0206 a AF022725A na chromozomu 7H, byly zpracovány programem MapManager, který pro markery RGH1aI1a, Bmag0021 a EBmag0794 vypočítal statisticky vysoce významnou vazbu s genem rezistence. Vazba s markerem RGH1aI1a v lokusu Mla potvrdila u zdroje rezistence PI466461 přítomnost genu v tomto lokusu. Nalezený gen je od tohoto markeru vzdálen 12 až 13 cM směrem k centromeře. Nový gen rezistence k padlí travnímu byl detekován na chromozomu 7H, v těsné vazbě s markerem Bmag0021. Vzdálenost genu od tohoto markeru je 3 cM směrem k centromeře.

Abstract:

The aim of this thesis was to study the genetic determination and localisation of new resistance genes in the new identified wild barley (Hordeum vulgare ssp. spontaneum) source of resistance to powdery mildew caused by the fungal pathogen Blumeria graminis f. sp. hordei. The genetic mapping of resistance genes was realised with SSR and CAPS markers in F2 generation that was created by cross between winter barley variety Tiffany and the source of resistance H. vulgare ssp. spontaneum PI466461, which is resistant to 22 European pathotypes of Bgh with wide range of virulence. The results of phytopathological studies revealed two resistance genes and their type of inheritance was evaluated as dominant and semidominant. The results of alelism test show, that one of these genes is localised in the Mla locus on chromosome 1HS. Lokalisation of the second gene was found by modified bulk segregant analysis (BSA) without DNA pooling. 34 SSR markers and the CAPS marker RGH1aI1a at the Mla lokus were tested by BSA. The linkage with a resistance gene was detected for markers RGH1aI1a, Bmac0213 and EBmac0501 on chromosome 1H, Bmag0500 on chromosome 6H and Bmag0021 on chromosome 7H. Mendelian segregation (i.e. a 1:2:1 ratio) was not confirmed for the marker Bmac0213 and this marker was excluded from further molecular analysis. Markers from the chromosomal regions where the linkage was detected by BSA were further tested for 97 F2 plants to identify the map positions of the looking resistance genes. The results of molecular analysis of individual F2 plants for markers RGH1aI1a, EBmac0501, Bmac0032, Bmac0063 and Bmac0090 on chromosome 1H, Bmag0500 and Bmac0316 on chromosome 6H and Bmag0021, EBmag0794, Bmag0206 and AF022725A on chromosome 7H were processed by the PC program MapManager which calculated the highly significant linkage with the resistance gene for markers RGH1aI1a at locus Mla on chromosome 1HS and for Bmag0021 and EBmag0794 on chromosome 7HS. The linkage with marker RGH1aI1a confirmed the presence of a resistence gene at locus Mla in the source of resistance PI466461. The gene is 12 to 13 cM far from this marker towards the centromere. The new gene for resistance to powdery mildew was detected on chromosome 7H in the linkage with marker Bmag0021. The gene is 3 cM far from this marker towards the centromere.
