Implementation and usage of CN-PAGE with gradient gel for separation of pigment-protein complexes

Abstract:

Tato diplomová práce je zaměřena na separaci pigment-proteinových komplexu (PPC) izolovaných z thylakoidních membrán (tBM) ječmene jarního pomocí bezbarvé nativní elektroforézy (CN-PAGE) s využitím gradientu gelu. Hlavním cílem bylo optimalizovat a standardizovat zavedenou metodiku CN-PAGE s gradientem gelu a použít jí ke zhodnocení působení světelného stresu na kompozici PPC izolovaných z thylakoidních membrán ječmene jarního. V teoretické cásti se zabývám popisem funkce a struktury hlavních PPC ? fotosystému II a fotosystému I a vlivem vysoké a nízké teploty a svetla na tyto základní struktury. Další dve kapitoly teoretické cásti tvorí strucná charakteristika chloroplastu a obecný popis elektroforézy a samotné metody CN-PAGE. V experimentální cásti je pojednáno o príprave extraktu tBM jecmene jarního a o stanovení obsahu chlorofylu a hlavní kapitolou je využití CN-PAGE pro separaci PPC z tBM vystavených zárení 100 ?mol m-2s-1 (LI) a 600 ?mol m-2s-1 (HI). Optimální separace PPC bylo dosaženo použitím kombinace detergentu DM a DOC pro prípravu vzorku i horního elektrodového pufru. Zóny byly zretelne oddeleny a došlo k separaci superkomplexu PSII-LHCII, PSI a dimeru PSII, monomeru PSII a cytochromu b6f, agregátu LHCII, LHCII v trimerním a monomerním stavu a volných pigmentu.

Abstract:

This diploma thesis has been focused on the separation of the pigment-protein complexes (PPC) insulated from thylakoid membranes (tBM) of the spring barley using the colorless native electrophoresis (CN-PAGE) with using the gradient of the gel. The main aim had been to optimize and standardize the implemented CN-PAGE methodology with the gradient of the gel and to use it for the evaluation of the effect of the light stress on the PPC composition insulated from thylakoid membranes of the spring barley. The theoretical part has been dealing with the description of the function and the structure of the main PPC ? the photo system II and the photo system I and with the influence of the high and low temperature and of the light on these basic structures. The other two sections of the theoretical part have been formed with the brief characteristic of the chloroplasts and the general description of the electrophoresis and the CN-PAGE methodology. The experimental part has been dealing with the tBM extract preparation of the spring barley and with the content determination of the chlorophyll and the main section has been the usage of CN-PAGE for the separation of the PPC from tBM exposed to the radiation of 100 ?mol m-2s-1 (LI) and of 600 ?mol m-2s-1 (HI). The optimal separation of PPC had been achieved using the combination of the DM and DOC detergents for the preparation of the samples and also the upper electrode buffer. The zones had been clearly separated and there had been separated the supercomplexes of PSII-LHCII, the PSI and PSII dimer, the PSII monomer and cytochrome b6f, the LHCII assembly and LHCII in the trimeric and monomeric state and the free pigments.
