The modulation of the cell differentiation of sarcoma cell lines with all-trans retinoic acid and LOX/COX inhibitors

Abstract:

Konvenční chemoterapie využívaná během minulých desetiletí v rámci léčby dětských onkologických onemocnění byla obvykle vysoce toxická a nespecifická, proto se v současnosti hledají jiné způsoby protinádorové léčby. Jednou z možností je indukovaná diferenciace transformovaných buněk do terminálních stádií. Jako induktory diferenciace se nejčastěji využívají retinoidy, především kyselina all-trans retinová (ATRA). Její nevýhodou je však zvýšená toxicita a krátkodobá intracelulární dostupnost. Tyto skutečnosti je možné ovlivnit pomocí kombinované aplikace ATRA s inhibitory lipoxygenáz (LOX) nebo cyklooxygenáz (COX), které zpomalují odbourávání ATRA v buňkách. Kombinace ATRA s LOX/COX inhibitory je využívaná v rámci metronomického protokolu COMBAT, který je určen pro dětské pacienty s relabujícími solidními nádory. Mechanismy působení této kombinované terapie však ještě nejsou zcela prozkoumány. Cílem předkládané práce proto bylo popsat a porovnat vliv samotné ATRA s účinky jejích kombinací s LOX/COX inhibitory na dvou osteosarkomových buněčných liniích (Saos-2 a OSA-1), s využitím kyseliny kávové jako inhibitoru 5-LOX, a celecoxibu jako inhibitoru COX-2. V rámci experimentů byly sledovány změny proliferace, četnost apoptózy a vliv na buněčnou diferenciaci buněk. U linie Saos-2 byla hypotéza o modulaci účinků ATRA potvrzena. Kombinace činidel dosáhly zvýšených účinků téměř ve všech analyzovaných procesech, kromě změn buněčného cyklu, kde se neprojevoval ani vliv samotných činidel. Obě kombinace činidel, ATRA/CA i ATRA/CX, dokázaly se stejnou efektivitou zvýšit antiproliferační účinek ATRA. Po aplikaci ATRA/CA se projevily větší změny na morfologické úrovni, kombinace ATRA/CX měla zase silnější cytotoxický potenciál a byla schopna zvýšit expresi diferenciačních markerů s vyšší účinností. U linie OSA-01 se také potvrdila očekávaná hypotéza. Výjimkou byla analýza buněčného cyklu a změn morfologie, kde se vliv samotných činidel ani jejich kombinací vůbec neprojevil. Na inhibici proliferace měla značně silnější účinky kombinace ATRA/CA, na indukci diferenciace bylo markantnější působení kombinace ATRA/CX. Výsledky prokazují, že modulace antineoplastických účinků ATRA pomocí LOX/COX inhibitorů na osteosarkomových buněčných liniích je úspěšná. Působení kombinací činidel je více efektivní, než působení činidel samotných.

Abstract:

In past, therapeutic approaches used for treating pediatric malignacies revealed high toxicity and low specificity. Therefore, new approaches in the therapy of cancer are needed. One possibility is induced differentiation of cancer cells into mature phenotypes. Retinoids, especially all-trans retinoic acid, are frequently used for the induction of the cell differentiation. However, they show substantial toxicity and are unstable. This can be overcame with the use of enzyme inhibitors, which participate in the catabolism of retinoids (5-lipoxygenase, cyclooxygenase-2). Combination of ATRA and LOX/COX inhibitors is used in the therapeutic regimen COMBAT (combined oral maintaining biodifferentiating and antiangiogenic therapy), which is intended for pediatric patients with relapsed solid tumors. Celecoxib is used as an inhibitor of cyclooxygenase-2 and caffeic acid as an inhibitor of 5-lipoxygenase. Molecular effects of this therapy have not been elluciated yet. The goal of diploma thesis is to compare the influence of all-trans retinoic acid alone compared to combined applictation of all-trans retinoic acid (ATRA) and LOX/COX inhibitors. Two osteosarcoma cell lines Saos-2 and OSA 01 were chosen. Proliferation, apoptosis and differentiation of control and treated cells were studied. The differences in the sensitivity of used cell lines were observed.In this diploma thesis, the hypothesis of the modulation of the differentiation effect of retinoids with LOX/COX inhibitors was confirmed in both studied cell lines. Saos-2 cells showed enhanced effects after agentscombinations in quite all analyzed procedures, except of changes in the cell cycle. Both combinations, ATRA/CA and ATRA/CX, were able to enhance the antiproliferative effect of ATRA alone. The applications of ATRA/CA caused more evident changes on the morphological level, while the combination ATRA/CX showed stronger cytotoxic potential and was able to increase the expression of used differentiation markers with higher efficiency then ATRA/CA. In contrast to Saos-2, OSA 01 cells demonstrated higher effect of the combination ATRA/CA on the inhibition of cell proliferation, while stronger differentiation potential has been shown using combination of ATRA/CX. Our results show the possibility of the modulation of antineoplastic effects of ATRA with LOX/COX inhibitors on osteosarcoma cell lines.
