Assessment of ecotoxicity of pure compounds and comlex environmental polutant mixtures with set of aquatic biotests

Abstract:

Obsahem této práce je praktické použití výluhových biotestů na testování jak přírodních směsných vzorku, tak i čistých chemických látek. Směsné přírodní materiály je před jejich dalším použitím vhodné zhodnotit z hlediska možných negativních účinků na cílovou lokalitu způsobených obsahem rizikových látek. Rovněž u nově vyvíjených látek je nutné otestováním zjistit jejich efekt na životní prostředí v případě možného úniku do prostředí. Základem práce je pojednání o třech různých skupinách látek a jejich následné praktické otestování baterií skládající se ze tří biotestů zahrnujícími tři základní trofické a organismální úrovně. Jedná se o testy s mikroskopickou řasou Pseudokirchnella subcapitata, sladkovodním korýšem Daphnia magna a mořskou luminiscenční bakterií Vibrio fischeri. Vzorky zahrnují sadu šesti sedimentů z kontaminovaných lokalit, kaly s vysokým obsahem minerálních látek ale kontaminované nadlimitním množstvím arsenu jako zástupce přírodních vzorků. Třetí bod zájmu této práce zahrnuje sadu nově vyvinutých termorezistentních výbušnin, které představují zástupce čistých látek. Výsledky naznačují, že vysoký obsah nebezpečných látek ještě nemusí způsobovat nebezpečnost matrice tyto látky obsahující. Určujícím faktorem je forma a biodostupnost nebezpečné látky. Rovněž bylo zjištěno, že určitými procesy lze měnit podíl a biodostupnost obsažených nebezpečných látek a ovlivnit tak vlastnosti matrice žádaným způsobem podle budoucího použití matrice. Výsledky testování výbušnin naznačují, že na základě příbuznosti látek se podobají i jejich toxické účinky. Na základě výsledků této práce se opětovně potvrzuje, že přístup testování pomocí biotestů je nenahraditelný, a že přináší jedinečné informace v procesu hodnocení toxicity jak přírodních vzorků, tak i čistých látek pomocí simulování skutečného působení látek na biotu

Abstract:

The aim of this thesis is practical using of aquatic biotests for testing natural mixture samples and pure chemical compounds. Complex environmental matrices as well as pure chemicals should be assessed before their future use as they can negatively affect the area of their application by adding contamination of toxic elements. The base part of this work is to provide basic information on three main groups of tested materials as well as basic information on three types of different biotests representing three trophic and organismal levels which were used to assess these matrices. These tests include the test with freshwater microscopic alga Pseudokirchnella subcapitata, the freshwater crustacean Daphnia magna and marine luminescent bacterium Vibrio fischeri. The studied materials includes: set of six potentially toxic sediments from contaminated localities, sludge samples with high content of arsenic as a environmental matrices. As example of pure compounds a set of eight thermoresistant explosives were chosen. The results in practical part of this work indicate that high content of toxic compounds does not mean directly high toxicity of the material. The crucial factor determining real toxicity of these toxic compounds is their form and bioavailability. There was also finding that several processes can change the portions and bioavailability of contained compounds so we can affect the matrix in the right way we require depending on the future manner of use of this material. Results of testing of explosives indicate that its chemical similarity causes their similar toxic effects. On the base of results presented in this thesis it has been proven that the approach using biotests is irreplaceable as it brings unique information usable in assessment of natural mixture toxicity as well as toxicity of pure chemicals via simulation of real exposure of organism to contaminant in natural conditions.
