The Biogenic Amine Production and Degradation in Brewery Microorganisms

Abstract:

Tato diplomová práce se zabývá produkcí a degradací biogenních aminů (BA) a po-lyaminů (PA) vybranými kmeny pivovarských kvasinek a laktobacilů. Izoláty byly získány od Ústavu pivovarského a sladařského v Praze. Byl proveden screening produkce a degradace biogenních aminů a polyaminů kvasinkami a laktobacily za podmínek in vitro, současně byl sledován vliv přídavku různých BA, PA a aminokyselin v několika variantách kultivačních médií. Stanovení bylo provedeno pomocí vysokoúčinné kapalinové chromatografie s UV detekcí s předkolonovou derivatizací dansylchloridem. Zástupci rodu Lactobacillus obecně disponují dekarboxylázovou aktivitou, a proto mohou představovat vážně riziko tvorby toxických množství BA/PA v pivu. To potvrzuje i analýzou zjištěná mohutná produkce tyraminu po kultivaci testovaných kmenů laktobacilů v živných médiích, která kolísala okolo 250 mg/l a místy dosahovala až 532 mg/l. Tyro-zindekarboxylázovou aktivitu vybraných kmenů laktobacilů značně podpořila i přítomnost tyrozinu coby prekurzoru tohoto BA. Nedostatek živin v nutričně chudých minerálních médiích však vedl k degradaci tyraminu testovanými kmeny laktobacilů, kdy jeho množství byly některé kmeny schopné snížit o téměř 120 mg/l. Na rozdíl od laktobacilů nepatří kvasinky mezi abundantní producenty. Byly zjiště-ny maximální přírůstky tyraminu do 23 mg/l, histaminu do 13 mg/l a putrescinu do 12 mg/l. Naopak nejvyšší úbytky se u tyraminu pohybovaly do 20 mg/l, u histaminu do 35 mg/l a u putrescinu do 12 mg/l. Pro potvrzení produkční/degradační schopnosti testovaných kmenů by bylo vhodné další experimenty navazující na tuto práci doplnit molekulárními metodami a stanovením po kultivaci kmenů v reálné matrici.

Abstract:

This thesis is dealing with the production and degradation of biogenic amines (BA) and polyamines (PA) by selected strains of brewing yeasts and lactobacilli. The isolates were provided by Research Institute of Brewing and Malting in Prague. The screening of production and degradation of BA and PA by brewing yeasts and lactobacilli was carried out at in vitro conditions, the influence of medium type and various supplements (BA, PA, amino acids) was observed. BA and PA were determined by high performance liquid chromatography with UV detection with precolumn derivatisation by dansyl chlori-de. Generally, the representatives of the genus Lactobacillus are able to decarboxylase amino acids to BA and PA. Therefore, their presence can be considered hazardous for the safety of beer. This statement was confirmed by a significant production of tyramine detec-ted after the cultivation of lactobacilli strains in nutrient media. Mentioned production fluctuated around 250 mg/l, at maximum 532 mg/l. The presence of tyrosine considerably contributed the tyrosinedecarboxylase activity of these microorganisms. However, the lack of nutrients in nutritionally poor mineral media led to the degradation of tyramine by Lactobacillus strains tested almost about 120 mg/l. Compared to lactobacilli, the yeasts are not classified as abundant producers of BA/PA. The maximum detected increments of tyramine was up to 23 mg/l, histamine up to 13 mg/l and putrescine up to 12 mg/l at maximum. By contrast, the highest decreases of tyramine were up to 20 mg/l, up to 34 mg/l of histamine and up to 12 mg/l of putrescine. To confirm the production/degradation capability of tested strains, it would be appropriate realize complementary molecular analysis and detection of BA/PA concentrations after the cultivation in real matrix in further experiments subsequent to this work.
