Typing and molecular epidemiology of human and porcine strains of Rotavirus A currently circulating in the Czech Republic

Abstract:

Rotaviry jsou příčinou akutního průjmového onemocnění u lidí i zvířat po celém světě, především u malých dětí do 5 let věku a u mladých jedinců mnoha živočišných druhů. U selat ve věku 4 týdnů, kdy jsou odstavena od prasnice, často dochází k poodstavovým průjmům, které mohou vést až k úhynu zvířat a především v chovech tak způsobují velké ekonomické ztráty. Kromě toho jsou rotaviry považovány za zoonotické agens, které mohou být přenášeny mezi člověkem a zvířaty. Zatím málo prostudovaným, ale zato významným rezervoárem rotavirů jsou volně žijící divoká prasata (Sus scrofa). V této práci bylo vyšetřeno 260 vzorků střevního obsahu divokých prasat odlovených na území ČR. Metodou reverzně transkripční PCR bylo detekováno 1,54 % vzorků pozitivních na RVA a 10,38 % na RVC. RVA pozitivní vzorky získané od divokých prasat byly spolu s pozitivními vzorky z trusu od prasat domácích dále charakterizovány sekvenací genomových segmentů kódujících významné povrchové proteiny VP4 a VP7 a dále proteiny VP6 a NSP4. Kombinace genotypů zjištěné u divokých prasat byly G9P[23]I5E1 a G1P[7]E1. U domácích prasat převládaly kombinace G11P[13], G5P[6] a G4P[6].

Abstract:

Rotaviruses are the cause of acute diarrhea in humans and animals all over the world, especially in young children under 5 years of age and in young ones of many animal species. Pigs at the age of 4 weeks after weaning often acquire rotaviral diarrhea, which can lead to death of animals and, in particular, in livestock, causes great economic losses. In addition, rotaviruses are considered to be zoonotic agents that can be transmitted between humans and animals. So far not studied enough, wild boars (Sus scrofa) may be a major reservoir of rotaviruses. In this thesis 260 samples of intestinal content of wild boars culled in the Czech Republic were examined. By reverse transcription PCR, 1.54% of samples positive for RVA and 10.38% for RVC were detected. RVA-positive samples obtained from wild boars and from domestic pigs were further characterized by sequencing of genomic segments encoding important VP4 and VP7 surface proteins and VP6 and NSP4 proteins. Combinations of genotypes found in wild boars were G9P[23]I5E1 and G1P[7]E1. In domestic pigs the predominant genotypes were G11P[13], G5P[6] and G4P[6].
