Optimalization of mesenchymal stem cell cultivation in bioreactor

Abstract:

Mezenchymální kmenové buňky (MSCs) se díky svému regeneračnímu potenciálu a imunomodulačním vlastnostem staly slibnou možností léčby různých onemocnění. MSCs lze snadno izolovat a rozmnožovat pomocí <em>in vitro</em> kultivace s plným využitím jejich schopnosti sebeobnovy. V současné době se MSCs pro klinické využití produkují pomocí konvenčních 2D kultur většinou v přítomnosti fetálního bovinního séra (FBS). Tyto metody však kvůli proměnlivým kultivačním podmínkám jako je špatně definované složení média a heterogenní kultivační prostředí nepředstavují pro MSCs ideální podmínky s využitím v regenerativní medicíně. Za tímto účelem se začaly rozvíjet možnosti 3D kultivace v bioreaktoru, které zároveň mají za cíl zvýšit produkci MSCs.<br> V rámci této práce byl nejdříve zkoumán vliv složek nahrazujících FBS v médiu (destičkový lyzát a KnockOut™ serum replacement) na množství a viabilitu MSCs. Hlavní část práce se zabývala optimalizací kultivace MSCs v bioreaktoru pomocí mikronosičů (MCs) Cultipsher-S a Cytodex 3. Nejdříve byla provedena kultivace pomocí MCs v menším objemu, kde byl optimalizován proces kvantifikace buněk na MCs a separace buněk od MCs. Následně byla zahájena kultivace v míchaném bioreaktoru (60 ml). Zkoumány byly především vlivy hypoxie a normoxie na celkové množství buněk a schopnost zachování specifických markerů při těchto podmínkách. Po dokončení kultivace se celkové množství MSCs nezvýšilo více než trojnásobně (2,80×). Je však potřeba brát v úvahu, že při 100% úspěšnosti separace MSCs od MCs by byl nárůst 4,26×. I přestože nárůst MSCs neodpovídal původním předpokladům, byla úspěšně otestována možnost kultivace MSCs v bioreaktoru pomocí MCs.

Abstract:

Mesenchymal stem cells (MSCs) have become a promising treatment option for various diseases due to their regenerative potential and immunomodulatory properties. MSCs can be easily isolated and expanded by <em>in vitro</em> culture, taking full advantage of their self-renewal capacity. Currently, MSCs for clinical use are primarily produced using conventional 2D cultures mostly in the presence of fetal bovine serum (FBS). However, due to variable culture conditions such as poorly defined media composition and heterogeneous culture environment, these methods do not represent ideal conditions for MSCs with applications in regenerative medicine. To address this, 3D bioreactor culture options have begun to be developed, aiming to increase MSCs production.<br> In this thesis, the effect of FBS replacement components in the medium (platelet lysate and KnockOut™ serum replacement) on the quantity and viability of MSCs was first investigated. The main part of this study was concerned with the optimization of MSCs cultivation in the bioreactor using microcarriers (MCs) Cultipsher-S and Cytodex 3. Initially, the culture using MCs was performed in a smaller volume, where the process of quantifying of cells on MCs and separating cells from MCs was optimized. Subsequently, cultivation in a stirred bioreactor (60 mL) was initiated. In particular, the effects of hypoxia and normoxia on total cell numbers and the ability to maintain specific markers under these conditions were investigated. After completion of cultivation, the total amount of MSCs did not increase more than threefold (2.80×). However, it should be taken into account that with 100% success rate of MSCs separation from MCs, the increase would be 4.26×. Even though the increase in MSCs did not match the original expectations, the possibility of culturing MSCs in the bioreactor using MCs was successfully tested.
