Modelling of separation by centrifugal partition chromatography (CPC)

Abstract:

Odstředivá rozdělovací chromatografie (CPC) je typ kapalinové chromatografie, jejíž stacionární i mobilní fáze jsou kapalné. Stacionární fáze je fixována v koloně (rotoru) pomocí odstředivé síly, zatímco mobilní fáze je pumpována skrz kolonu. Látky, procházejíce opakovaným rozdělováním mezi oběma fázemi, jsou separovány podle svých rozdělovacích koeficientů. Jelikož lze použít velkou paletu systémů rozpouštědel, je CPC velmi univerzální metoda, umožňující separaci širokého spektra látek (od úrovně proteinů po malé organické molekuly). V bakalářské práci je studován vliv operačních podmínek, jako je rychlost rotace rotoru nebo rychlost průtoku mobilní fáze, na separaci. Cílem práce je analyzovat závislost různých parametrů ovlivňujících separaci na operačních podmínkách. V práci jsou prezentovány vytvořené modely a s využitím statistických nástrojů je hodnocena jejich kvalita. Dílčím výstupem této práce je i softwarový nástroj, umožňující simulaci separace na CPC. Data pro tuto práci byla poskytnuta výzkumnou skupinou Karine Faure, PhD (ISA, Lyon, Francie).

Abstract:

A centrifugal partition chromatography (CPC) is a type of liquid chromatography, of which both, the stationary and mobile phase, are liquid. The stationary phase is retained in the column (rotor) by the centrifugal force, while the mobile phase is pumped through the column. The solutes undergoing the process of continuous partitioning between the phases are separated accordingly to their partitioning coefficients. CPC is a very versatile technique allowing the user to separate a wide range of compounds (from the level of proteins to small organic molecules) as a large variety of solvent systems can be used. In this bachelor’s degree project, the influence of operating conditions, such as the flow-rate and rotation speed, on the separation is studied. The objective of the work is to analyse the dependence of the parameters influencing the separation on the operating conditions. In the work, the created models are presented and assessed with the use of statistical tools. The output of this work is a software tool allowing the simulation of a separation on CPC. The data for this study were provided by the research group of Dr. Karine Faure (ISA, Lyon, France).
