The response of bee immune system to viral infections

Abstract:

V této bakalářské práci jsou shrnuty dosavadní vědecké poznatky o imunitních reakcích včely medonosné (Apis melifera) proti virovým infekcím. Včela medonosná má nezastupitelnou roli jako opylovač mnoha hospodářsky významných rostlin. V posledních letech byl zaznamenán úbytek včelích kolonií, na nichž se podílí mnoho faktorů, mezi které patří i viry. Včela se proti virovým infekcím brání vrozenou imunitou, kterou lze rozdělit na individuální a sociální imunitu. Individuální imunitu lze pak dále rozdělit na humorální a buněčnou imunitu. Nezastupitelnou složkou v boji proti virům je RNA interference, pomocí které je imunitní systém včely schopen cílit přímo na virovou genetickou informaci a blokovat ji. V práci jsou také popsány imunitní dráhy, které samy o sobě nesníží množství viru. Nicméně, regulace exprese genů kódujících efektorové molekuly má vliv na obranu proti virům. Významnými efektorovými molekulami jsou antimikrobiální peptidy, ačkoliv mechanismus jejich zapojení do boje proti virům není uspokojivě objasněn. Přesto se ukazuje, že jejich role v antivirální obraně je klíčová. U procesu melanizace, a také u skupiny proteinů teplotního šoku a proteinů obsahující thioesterovou skupinu byla taktéž prokázána jejich role v antivirální obraně. Poněkud komplikované je určit, zda buněčná imunita přispívá ke snížení virové infekce ve včelích koloniích. Přesto, že se u jiných druhů hmyzu a modelových organismů ukazuje, že procesy jako je fagocytóza a nodulace se podílejí na obraně proti virům, u včely medonosné nejsou tyto procesy, v souvislosti s obranou proti virům, dostatečně prozkoumané, aby se dalo toto tvrzení podpořit. Nedílnou součástí imunitního systému včely medonosné je sociální imunita, která zahrnuje koordinované reakce jednotlivých včel využívané na obranu včelstva proti patogenům a její zapojení v antivirální obraně je v práci také diskutováno. Konkrétně proti virům využívají včely mechanismy jako je grooming neboli čištění, hygienické chování a sběr pryskyřice, které mohou přispět ke snížení virové nálože ve včelstvu.

Abstract:

This bachelor’s thesis summarizes the current scientific knowledge about the immune responses of honeybee (Apis mellifera) against viral infections. The honeybee has an irreplaceable role as a pollinator of many economically important plants. In recent years, there has been a decline in bee colonies involving many factors, including viruses. The bee defends itself against viral infections by innate immunity, which can be divided into individual and social immunity. Individual immunity can then be further divided into humoral and cellular immunity. An irreplaceable component in the fight against viruses is RNA interference, by which the bee’s immune system can target and block viral genetic information directly. The thesis also describes immune pathways that do not in themselves reduce the amount of virus. However, regulation of the expression of genes encoding effector molecules has an effect on defence against viruses. Important effector molecules are antimicrobial peptides, although the mechanism of their involvement in the fight against viruses has not been satisfactorily elucidated. Nevertheless, their role in antiviral defence is proving to be crucial. The role in antiviral defence has also been demonstrated in the melanization process, as well as in the family of heat shock proteins and the thioester group-containing proteins. It is complicated to determine whether cellular immunity contributes to the reduction of viral infection in bee colonies. Although in other insect species and model organisms have been shown to contribute to virus defence in processes such as phagocytosis and nodulation. In honeybees these processes have not been sufficiently investigated in relation to virus defence to support this claim. A significant part of the immune system of the honeybee is social immunity, which includes coordinated reactions of individual bees used to defend bee colonies against pathogens and its involvement in antiviral defence is also discussed in the thesis. In particular, against viruses bees use mechanisms such as grooming, hygienic behaviour and resin collection, which can help reduce the viral load in the hive.
