Target-random mutagenesis of RSL lectin

Abstract:

This thesis is focused on target-random mutagenesis of the RSL lectin, which consists of creating a random mutation in preselected spots: Ala40, because it participates on binding saccharides and can affect the selectivity and affinity, Gly39, because it can keep binding pocket shape and Asp32, because it increases potential of hydrogen bonding. The aim of this thesis is optimization of mutation PCR and high-throughput screening methods. Besides a literary review, the bachelor thesis contains a description of the used methods and results, which summarize the processes of the target-random mutagenesis and testing of the expression and purification of RSL protein with six oligohistidine tags for his possible application in high-throughput screening.

Abstract:

Tato práce se zaměřuje na cíleně-náhodnou mutagenezi lektinu RSL, která spočívá ve vytvoření náhodné mutace na dopředu zvoleném místě, a to Ala40, protože se podílí na vazbě sacharidů a zodpovídá za jejich selektivitu a aktivitu, Gly39, protože drží strukturu vazebního místa a Asp32, protože zvyšuje potenciál vodíkové vazby. Práce je primárně zaměřena na optimalizaci mutační PCR a některých metod high-throughput screeningu. Bakalářská práce obsahuje kromě zpracované literární rešerše také popis užívaných metod a výsledků, které sumarizují postupy cíleně-náhodné mutageneze a testování exprese a purifikace proteinu RSL s šesti oligohistidinovými kotvami pro jeho možné uplatnění při high-throughput screeningu.
