Kinetics of tramadol hydrochloride release from lipophilic matrix tablets

Abstract:

Tato diplomová práce se zabývá kinetikou uvolňování tramadol hydrochloridu (jakožto účinné látky) z lipofilních matricových tablet s různým obsahem retardující složky glycerol-dibehenát (Compritol?). Byl studován vliv teploty na rychlost uvolňování této účinné látky z matricových tablet. Lipofilní matricové tablety byly připraveny metodou přímého lisování. Následně byly provedeny disoluční testy v kyselém disolučním médiu o pH 1,2 při teplotách 35, 37 a 40 °C. Získané disoluční profily byly vyhodnoceny pomocí matematických modelů: kinetický model 1. řádu a model Kormeyer-Peppas. Bylo zjištěno, že se zvyšujícím se obsahem retardující složky se snižuje vliv teploty na rychlost uvolňování tramadol hydrochloridu.

Abstract:

This master thesis is concerned with the kinetics of tramadol hydrochloride (as active substance) release from lipophilic matrix tablets with different contents of the retarding component glycerole-dibehenate (Compritol?). The effect of temperature on the release rate of this active substance from the tablets was studied. Lipophilic matrix tablets were prepared by direct compression. Subsequently, dissolution tests were performed in an acidic dissolution medium of pH 1.2 at temperatures of 35, 37 and 40 °C. The obtained dissolution profiles were evaluated using mathematical models: the 1st order kinetic model and the Kormeyer-Peppas model. It was concluded that with increasing amount of glycerole-dibehenate in the tablet, the effect of temperature on the release rate of tramadol hydrochloride decreases.
