Ecotoxicological biotests on embryolarval stages of aquatic organisms - questions of the considerate handling of the model organisms

Abstract:

Funkce ekotoxikologických biotestů in vivo je i přes současné trendy, směřující spíše k in vitro a in silico testování, stále nenahraditelná. Důležitou podmínkou pro aplikaci testů in vivo v moderní ekotoxikologické praxi je dodržení pravidel maximálně šetrného zacházení s modelovými organismy (vhodné podmínky chovu, šetrná manipulace, omezení či úplné vyloučení bolestivých podnětů během experimentu pomocí užití šetrných operačních metod či aplikace anestézie). Tato práce se zabývá možnými způsoby humánního ukončování testu s embryolarválními stádii drápatky vodní (FETAX). Shrnuje a porovnává aplikaci různých veterinárních anestetik za účelem snížení stresu při fixaci embryí drápatky roztokem formaldehydu, která je zavedena jako standardní způsob ukončování testu v metodice ASTM. V praktické části práce byla hodnocena efektivita dvou alternativních látek (tricain MS 222 a hřebíčkový olej) pro anestetizaci 96 hodin starých embryí drápatek z vlastního odchovu. Aplikace anestézie byla vyhodnocena jako důležitý faktor pro eliminaci stresu u testovaných jedinců a zvýšení humánnosti testu.

Abstract:

Even though the current trends in ecotoxicology tends to the use of in vitro and in silico testing, in vivo assays are still irreplaceable. However, it is important to apply the rules of maximally considerate handling of the model organisms (aproppriate breeding conditions, considerate treatment, restriction or absolute elimination of painful stimuli while performing the experiment by means of considerate surgical methods or application of anesthesia) while using in vitro bioassays in modern ecotoxicology. This work focuses on possible ways of humane ending of the Frog Embryo Teratogenesis Assay Xenopus (FETAX). It summarizes and compares the application of various veterinary anesthetics in order to lower stress in embryos of African clawed frogs while fixating them with formaldehyde solution, which is adopted as standardized way of ending the assay in ASTM methodology. In the practical part of the work, effectivity of two alternative compounds (tricaine MS 222 and clove oil) for anesthetisation of clawed frogs was evaluated. Application of the anesthetics was rated as an important factor for elimination of stress in tested individuals and increasing humanity of the assay.
