Characterization of the C-terminal domain of Nab3 protein

Abstract:

V této práci jsem provedl strukturní studii C-term domény proteinu Nab3 metodami nukleární magnetické rezonance (NMR) a rozptylu Roentgenova záření pod malými úhly (SAXS). Provedl jsem přiřazení chemických posunů pro páteřní atomy a atomy postranních řetězců v C-term doméně. Výsledky jsem pak použil pro strukturní výpočet založený na měkkých vazebných podmínkách odvozených z NOESY spekter. V průběhu studie jsme na základě SAXS dat zjistili, že C-term doména tvoří v roztoku homodimer. Za účelem potvrzení výsledků ze SAXS jsme provedly měření NMR relaxací, které potvrdily dimer. Ve světle těchto zjištění jsem se pokusil na základě struktury monomeru vypočítat nejpravděpodobnější strukturu dimeru C-term domény, kterou v závěru této práce prezentuji.

Abstract:

In this thesis I have conducted a structural study of a C-term domain of a protein Nab3 by methods of the Nuclear Magnetic Resonace (NMR) and the Small Angle X-ray Scattering (SAXS). I have done a backbone and a side chain assignment of a nuclear resonance frequencies of the C-term domain and used it to calculate a structure of the C-term domain based on a NOESY spectra. We also found, based on a SAXS data, that the C-term domain probably form a dimer in a solution. In order to verify this speculation we have measured an NMR relaxation which confirm the dimer state of the C-term domain. Based on these data I have calculated a potential structure of the dimer based on a monomer structures and present it in my thesis.
