Study of long non-coding RNAs in breast cancer

Abstract:

Breast cancer possess highest incidence among cancers in women worldwide. Current therapy of this disease is insufficient, especially in aggressive subtypes like triple-negative or luminal-B breast cancer. An important assumption for successful therapy of breast cancer is early detection and therefore there is a need for identification of new diagnostic and prognostic markers. High potential in this field indicate long non-coding RNAs (lncRNAs), which are frequently deregulated in tumors, where they act as oncogenes or as tumor suppressors. One of the studied oncogenic lncRNAs is MALAT-1, which is over-expressed also in breast cancer. The aim of the thesis was to determine the expression level of MALAT-1 in breast cancer cell lines, silence MALAT-1 in selected cell lines and monitor the effect of estrogen on the expression of MALAT-1. Consequently, MALAT-1 effects on proliferation, apoptosis, migration and clonogenicity were analyzed in vitro and potential synergistic functioning of MALAT -1 silencing and estradiol treatment was evaluated. The results of this work suggest that MALAT-1 is involved in the process of clonogenicity, which is seems to be further potentiated by estradiol. Involvement of MALAT-1 in proliferation, apoptosis and migration was not significant

Abstract:

Karcinom prsu patří mezi nádorové onemocnění s nejvyšší incidencí u žen na celém světě. Léčba tohoto onemocnění je nedostačující a to především u agresivních typů, jakými jsou triple-negativní a luminal B karcinomy. Důležitým předpokladem úspěšné léčby je včasná diagnostika, a proto je nezbytná identifikace nových diagnostických a prognostických markerů. Potenciál v této sféře mají dlouhé nekódující RNA (lncRNA), které jsou často deregulovány u nádorových onemocnění, kde působí jako onkogeny nebo jako nádorové supresory. Jednou z onkogenních lncRNA je MALAT-1, jejíž zvýšená exprese byla popsána také u karcinomu prsu. Cílem práce bylo stanovit hladinu MALAT-1 v buněčných liniích karcinomu prsu, ve vybraných liniích utlumit hladinu MALAT-1 a sledovat působení estradiolu na jeho expresi. Následně in vitro funkčně charakterizovat efekty MALAT-1 na proliferaci, apoptózu, migraci a klonogenicitu a ověřit možné synergistické působení MALAT-1 a estradiolu. Výsledky této práce nasvědčují, že MALAT-1 je zapojený do procesu klonogenicity a zřejmě je toto působení dále potencované estradiolem.
