Abstract:

Moje disertační práce se skládá ze dvou částí, (i) teoretické části shrnující problematiku rostlinných telomer, telomerázy a epigenetické regulace telomer, (ii) seznamu 9 publikací. Telomery jsou nukleoproteinové struktury na koncích eukaryotických chromozomů, potřebné k odlišení přirozených konců chromozomů od dvouřetězcových zlomů DNA. Dále zabraňují ztrátě kódujících sekvencí DNA během replikace. Telomery živočišných buněk se v průběhu života buňky zkracují, u rostlin je délka telomer stabilní. Proto bylo poměrně překvapivé zjištění, že telomery se prodlužují v kalusových kulturách Silene latifolia a Arabidopsis thaliana. Na druhou stranu, v kalusech tabáku (Nicotiana tabacum) se délky telomer neměnily. V naší práci jsme se zaměřili na dynamiku délky telomer u dlouhodobě propagovaných kalusů a rostlin z těchto kalusů regenerovaných. Pozorovali jsme postupné prodlužování telomer u kalusových kultur odvozených z klíčních rostlin tří ekotypů A. thaliana (Columbia, Wassilevskija, Landsberg erecta); u kalusových kultur N. tabacum a N. sylvestris byly telomery, v souladu s předchozím pozorováním stabilní. V kalusech byl dle očekávání aktivní enzym telomeráza (ribonukleoproteinový komplex schopný prodlužovat telomery), analýzy však neprokázaly korelaci délek telomer se změnami aktivity telomerázy. Stejně tak nebyla pozorována změna dynamiky délek telomer v kalusech založených z klíčních rostlin s poruchou funkce genů kódujících epigeneticky významné proteiny. Je tedy pravděpodobné, že důvodem porušení homeostázy telomer pozorované během dlouhodobé kultivace kalusů A. thaliana je kombinace faktorů spojených s procesy dediferenciace a dlouhodobé in vitro kultivace. Konce chromozomů jsou tvořeny relativně konzervovanou repetitivní sekvencí, u většiny rostlin je to v C-bohatém vlákně sekvence 3´-AAATCCC-5´. Všechny cytosiny této telomerové repetice mohou být cílem de novo DNA methyltransferáz. Methylaci telomer a methylaci DNA obecně lze studovat s využitím biochemických nebo genetických přístupů. Biochemický přístup využívá např. hypomethylační činidla ovlivňující aktivitu příslušných methyltransferáz (zebularin, DHPA); genetický přístup využívá inzerce T-DNA do genů kódujících proteiny klíčové pro udržení stabilního methylačního obrazu. U rostlin A. thaliana klíčících v přítomnosti zebularinu nebo DHPA a u methylačních mutantů jsme pozorovali pokles hladiny methylace cytosinů lokalizovaných v telomerových repetitivních sekvencích.U hypomethylovaných rostlin se významně zkrátily telomery a tento fenotyp se přenesl i do dalších generací rostlin, které už nebyly zebularinu ani DHPA vystavené. Zkracování telomer po opakovaném působení hypometylační látky v časných fázích vývoje rostliny má své limity; telomery v dalších generacích rostlin ovlivněných zebularinem během klíčení se už dále nezkracovaly. Dynamika délek telomer v hypomethylovaných rostlinách byla studována i ve vztahu k transkripční aktivaci transpozabilních elementů (TE), která byla taktéž pozorována jako důsledek působení zebularinu. Zajímavé bylo, že transkripce TE se vrátila na původní úroveň okamžitě po ukončení působení zebularinu, zatímco návrat telomer na původní délku byl pozvolný. To ukazuje, že rostliny mají vybudované sofistikované a diferencované mechanismy, které chrání genom před potenciálně škodlivými důsledky aktivace TEs a zkrácení telomer. Data získaná během této disertační práce přispěla k poznání dalších mechanismů, které jsou zapojeny do komplexního procesu regulace délky telomer rostlin. Abstract:

My Ph.D. thesis consists of two parts, (i) the theoretical part summarizing the issues of plant telomeres, telomerase and epigenetic regulation of telomeres, (ii) list of nine publications. Telomeres are nucleoprotein structures at the ends of eukaryotic chromosomes essential for distinguishing natural chromosome ends from double stranded DNA breaks, and protection of coding DNA loss during replication. Telomeres of animal cells shorten during the cell life; in plants, telomere lengths are maintained throughout their life. Therefore, detection of elongated telomeres detected in Silene latifolia and Arabidopsis thaliana calli was rather surprising. On the other hand, in tobacco callus cultures, lengths of telomeres were stable. In our work, we were interested in telomere lengths dynamics in long-term propagated calli and regenerated plants. We observed gradual telomere elongation in calli derived from seedlings of three A. thaliana ecotypes (Columbia, Wassilevskija, Landsberg erecta), while in tabacum and N. sylvestris calli, lengths of telomeres were stable, in accordance with previous observations. As expected, telomerase enzyme (ribonucleoprotein complex able to extend 3´-terminus of the G-rich strand of telomeres) was active in callus cultures, but there was no correlation between telomere elongation and changes of telomerase activity. Dynamics of telomere lengths changes was not affected in calli originated from seedling with loss of function of genes encoding epigenetically important proteins. Thus, it is probable, that disruption of telomere homeostasis in A. thaliana calli arises from the interplay of multiple factors, as a part of reprogramming of plant cells to long-term culture conditions. The ends of chromosomes are formed by relatively conserved repetitive sequences, 3´-AAATCCC-5´ in the C-rich telomeric strand in most of plants. All cytosines in this telomeric repeat can be targets for de novo DNA methyltransferases. There are two basic approaches for the study of telomere methylation, biochemical and genetic. In the biochemical approach, chemicals, e.g., hypomethylating agents affecting the activity of methyltransferases (zebularine, DHPA) are utilized; in the genetic approach, transgenic plants with T-DNA inserted into genes coding for proteins crucial for the maintenance of the stable methylation pattern are studied. In our experiments, the decrease of telomeric cytosines methylation was observed in A. thaliana plants germinated in the presence of zebularine or DHPA, and in methylation mutant plants. Telomeres were significantly shortened in hypomethylated plants and the short telomere phenotype was transmitted into the next plant generations growing without the hypomethylation drug exposure. It was further demonstrated, that telomere shortening after repeated drug exposures during early plant development was limited; no progression in telomere shortening was observed in subsequent generations of plants exposed to zebularine. Dynamics of telomere lengths changes in hypomethylated plants was correlated with transcriptional activation of transposable elements (TEs), which was also detected as a consequence of the zebularine exposure. Interestingly, TEs transcription dropped sharply immediately after finishing the zebularine treatment while telomere lengths recovery was gradual. This observation points to sophisticated and distinguished mechanisms protecting plant genome from the detrimental consequences of TEs activation and telomere shortening. Data presented in this Ph.D. thesis contributed to the elucidation of other mechanisms involved in the complex process of plant telomere lengths regulation. As results in this field obtained in analyses of different plant and animal representatives are often different, study of other plants and animals with canonical and non-canonical telomeric sequences is essential for understanding of both general and species-specific processes involved in the telomere lengths maint
