There are monovalent and divalent cations naturally localised inside of the cells, which could influence interaction between protein and DNA. In this thesis, we were studying the impact of ions to binding properties of protein IFI16 and his domains. Uvnitř buňky se přirozeně vyskytují jednomocné a dvoumocné kationty, které mohou ovlivňovat interakce mezi proteinem a DNA. V této práci jsme zkoumali vliv iontů na vazebné vlastnosti proteinu IFI16 a jeho domén. Pomocí gelové retardační analýzy a fluorescenční anizotropie jsme zjistili, že vliv zkoumaných kationtů se lišil při vazbě proteinu na lineární a na superhelikální DNA. Také jsme pozorovali …more
