Identification of forensically important insect species based on DNA analyzes

Abstract:

Nekrofágní hmyz představuje významnou roli v kriminalistické praxi. Ve forenzní entomologii se vzorky hmyzu, které se nachází na lidských mrtvolách nebo na místě činu, používají na odhad posmrtného intervalu (PMI). Významní jedinci kolonizují ostatky v sukcesních vlnách, je proto důležité znát posloupnost hmyzu v jednotlivých vlnách pro přesnější odhad PMI. Jejich výskyt je ale ovlivňován i klimatickými podmínkami, především teplotou. Nejdůležitější zástupci forenzně významného hmyzu jsou z řádů Diptera a Coleoptera. Je tedy důležité jednotlivé druhy hmyzu správně identifikovat, díky odhadu PMI může být následně úspěšně vyhodnocen trestný čin. Na morfologickou identifikaci se využívají morfologické klíče, ale taková identifikace může být obtížná kvůli podobné morfologii zejména ve stadiu larvy a kukly. Pro molekulární identifikaci, která je založená na analýze DNA, je nejvíce používanou metodou DNA barcoding, ale i RFLP, RAPD, AFLP a fylogenetická analýza. Jako molekulární markery se využívají jaderné i mitochondriální geny, především COI.

Abstract:

Necrophagous insects play an important role in forensic practice. In forensic entomology, insect samples found on human corpses or at the crime scene are used to estimate the postmortem interval (PMI). Significant individuals colonize the remains in succession waves, so it is important to know the sequence of insects in each wave for a more accurate estimate of PMI. However, their occurrence is also influenced by climatic conditions, especially temperature. The most important representatives of forensically important insects are from the orders Diptera and Coleoptera. It is therefore important to correctly identify the individual insect species, thanks to the PMI estimate, the crime can then be successfully evaluated. Morphological keys are used for morphological identification, but such identification can be difficult to influence similar morphologies, especially at the larval and pupal stages. For molecular identification, which is based on DNA analysis, the most used method is DNA barcoding, but also RFLP, RAPD, AFLP and phylogenetic analysis. Nuclear and mitochondrial genes, especially COI, are used as molecular markers.
