Phytochemical analysis of blackberry fruit

Abstract:

Plody ostružiníků jsou hojně konzumovány jako ovoce a byl v nich popsán obsah několika bioaktivních sloučenin, které mohou mít pozitivní vliv na lidské zdraví. Popisu ostružiníku, jeho plodů a nejčastěji analyzovaných látek při jejich chemické charakterizaci je věnována teoretická část této práce. V experimentální části jsou popsány provedené analýzy vybraných genotypů ostružiníku. Byly provedeny experimenty stanovující mastné kyseliny, cukry, vitaminy, fenolické a flavonoidní látky, antioxidační aktivitu a těkavé látky. Využity byly metody chromatografické, spektrometrické a spektrofotometrické. Dominantní mastnou kyselinou ve všech vzorcích byla kyselina linolová, následovala kyselina olejová, palmitová a stearová. Z jednoduchých cukrů byly stanoveny glukosa a fruktosa. Z vitaminů vykazoval nejvyšší obsah vitamin C, B3, B5, v menším množství pak další vitaminy skupiny B a -tokoferol. Z fenolických látek byl nejzastoupenější katechin, kyselina gallová a 2-O-B-D-glukosid kyseliny salicylové. Analýzy celkového obsahu fenolických látek a antioxidační aktivity vykazovaly silnou korelaci. Bylo identifikováno 25 těkavých látek, jejich zastoupení ve vzorcích bylo variabilní. Práce podává chemickou charakterizaci plodů daných genotypů ostružiníku a jejich vzájemné porovnání. Zejména byly pozorovány odlišnosti mezi ostružinami kultivarů a divokých druhů, a to konkrétně v obsahu cukrů, vitaminů skupin B a C, celkovém obsahu fenolických látek a antioxidační aktivitě.

Abstract:

The blackberries are widely consumed fruit and their content of several bioactive compounds, that can have a positive effect on human health, have been described. The theoretical part of this bachelor work presents the description of Rubus plants, their fruits, and the most frequented analytes in their chemical characterization. The experimental part describes performed analyses of selected Rubus genotypes. The determination of fatty acids, sugars, vitamins, phenolics and flavonoids, antioxidant activity and volatiles were performed using chromatographic, spectrometric and spectrophotometric methods. The dominant fatty acid in all samples was linoleic acid, followed by oleic, palmitic and stearic acids. Determined monosaccharides were glucose and fructose. Most abundant vitamins were C, B3, B5, followed by other B vitamins and -tocopherol. From phenolics, most common were catechin, gallic acid and salicylic acid 2-O-B-D-glucoside. The total phenolic content and antioxidant activity were strongly correlated. Twenty-five volatiles were identified among the samples, their representation was variable. This work provides chemical characterization of selected blackberry genotypes and their mutual comparsion. Especially, differences between cultivated and wild blackberries were spotted, in particular in contents of sugars, vitamins B and C, total phenolics and antioxidant activity.
