The effect of chemotherapy on the oncogenic potential of tumor cells grown in 3D cultures

Abstract:

Perifosin je nový chemoterapeutický přípravek, inhibitor signální dráhy PI3K/AKT, která bývá často nadměrně aktivována v rozličných typech rakoviny, včetně kolorektálního karcinomu (CRC). Tato dráha podporuje buněčný růst a přežívání, tudíž představuje specifický molekulární cíl pro protinádorovou léčbu. Ačkoli preklinické studie ukazovaly slibné výsledky perifosinové efektivity, klinické studie fáze III nepotvrdily jeho benefit pro léčbu pacientů s diagnózou kolorektálního karcinomu. V této studii byl pro posouzení cytotoxicity perifosinu v lidských solidních nádorech aplikován trojrozměrný (3D) in vitro kultivační systém nazývaný mnohobuněčné sféroidy (MCS). MCS mají přirozeně několik vlastností podobných nádorům, jako gradient živin, kyslíku, pH a proliferace a taktéž napodobují bariéry pro penetraci terapeutik dovnitř tkáně. Poskytují proto lepší model nádorového mikroprostředí než dvojrozměrné (2D) kultury. Cytotoxicita perifosinu byla sledována na 3D modelu buněčné linie HCT 116 odvozené z kolorektálního karcinomu. Ke zkoumání cytotoxických účinků byly použity testy MTT a ATP a fluorescenční barvení calcein AM/propidium jodid. Vliv perifosinu na indukci apoptózy byl sledován pomocí imunoblotingu a imunohistochemie s využitím apoptotického markeru štěpené kaspázy 8 a proteinu PARP. Výsledky naznačují, že protinádorové účinky perifosinu jsou závislé na vlastnostech prostředí, ve kterém se nádor vyskytuje, zvláště na hodnotě pH. V acidických podmínkách typických pro nádorové mikroprostředí i lumen tlustého střeva účinnost perifosinu rapidně klesá.

Abstract:

Perifosine is a novel chemotherapeutic agent, an inhibitor of PI3K/AKT signalling pathway, which is often upregulated in different types of cancer, including the colorectal carcinoma (CRC). This pathway promotes cell growth and survival and therefore represents a specific molecular target for anticancer treatment. Although the preclinical studies showed promising results of perifosine effectivity, the phase III clinical trial did not confirm its survival benefit for the treatment of patients with colorectal carcinoma. To assess the cytotoxicity of perifosine in human solid tumours, the three-dimensional (3D) in vitro culture system called multicellular spheroids (MCS) was applied. MCSs naturally possess several tumour-like characteristics, such as the gradient of nutrients, oxygen, pH and proliferation and also mimic barriers for penetration of therapeutics inside the tissue. Therefore, they provide a better simulation of tumour microenvironment than two-dimensional (2D) monolayers. The cytotoxicity of perifosine was studied using a 3D model of the cell line HCT 116 derived from colorectal carcinoma. To examine the cytotoxic effects, the MTT and ATP assays and the fluorescent staining by calcein-AM/propidium iodide were applied. The effect of perifosine on the apoptosis induction was examined by protein immunoblotting and immunohistochemistry using the apoptotic marker cleaved caspase 8 and PARP protein. Overall results suggest that the anti-tumour effects of perifosine are environment-dependent and particularly dependent on the pH value. In the acidic conditions, which are typical for the tumour microenvironment and the lumen of the colon, the efficacy of perifosine decreases rapidly.
