Screening of Non-starter Bacteria Isolated from Food to Degradation of Biogenic Amines

Abstract:

Cílem této práce bylo ověření schopnosti degradace biogenních aminů vybranými bakteriemi, jelikož bakterie s aktivitou aminooxidáz jsou v poslední době v popředí pozornosti z důvodu možnosti snižování koncentrace biogenních aminů v potravinách. Teoretická část líčí biogenní aminy, popisuje jejich vznik, produkci bakteriemi, výskyt v potravinách a možnosti jejich mikrobiální degradace. Praktická část této práce se zabývala ověřením schopnosti degradace biogenních aminů vybranými kmeny a poté byl chromatograficky stanoven úbytek biogenních aminů v médiu Nutrient Broth s biogenními aminy a 1/2 médiu Nutrient Broth s biogenními aminy, při 24 a 48 hodinové kultivaci pomocí vysokoúčinné kapalinové chromatografie po předchozím převedení na deriváty, které bylo možno detekovat v UV oblasti. K významnému úbytku u všech sledovaných biogenních aminů došlo kmenem Citrobacter gillenii B58 a kmenem Citrobacter gillenii B59. Dobrou degradační aktivitou disponoval také kmen Enterococcus faecium B51.

Abstract:

The aim of this work was to verify the biodegradability of biogenic amines by selected bacteria, because bacteria with amino acid activity have been at the forefront of attention due to the possibility of reducing the concentration of biogenic amines in food. The theoretical part describes biogenic amines, their origin, bacterial production, occurrence in food and possibilities of their microbial degradation. The practical part of this work deals with verification of biogenic amine biodegradability by selected strains and then chromatographically determines the loss of biogenic amines in Nutrient Broth medium with biogenic amines and 1/2 medium Nutrient Broth with biogenic amines at 24 and 48 hours cultivation by high performance liquid chromatography after previous conversion to derivatives that can be detected in the UV area. Significant loss of all biogenic amines observed occurred in Citrobacter gillenii B58 strain and Citrobacter gillenii B59 strain. The strain Enterococcus faecium B51 had also a good degradation activity.
