Abstract:

Tato práce se zabývá instrumentálními a konstrukčními principy, které jsou nezbytné pro vytvoření systému určeného k analýze komplexních vzorků biologické povahy. Studium a vývoj mikrofluidických zařízení byl zaměřen na spojení hmotnostní spektrometrie, která slouží jako detekční prvek, s elektromigračními technikami, které plní funkci separační. Primární oblastí pro využití tohoto systému byla proteomická analýza, při níž byla využita skupina peptidů a proteinů pro testování a následnou optimalizaci. Jedním z hlavních cílů tohoto vývoje bylo take nalezení vhodného materiálu a dosažení vysoké citlivosti. V případě materiálů je nutné vyhnout se polymerům s příměsemi změkčovadel a látek, které způsobují potlačení signálu při ionizaci elektrosprejem. Další cíl byl směřován k celkové miniaturizaci systému a možností pracovat s malými objemy vzorku, při zachování spolehlivosti a kompaktnosti zařízení.

Abstract:

This thesis explores the instrumentation principles and construction background of hyphenated techniques which are required for analysis of complex samples in the field of bioanalytical chemistry. Study and development of microfluidic devices was entirely focused on coupling of mass spectrometry with electromigration separation techniques. Part of the efforts was directed to the area of proteomic analysis, where peptides and proteins were used as model samples for testing and optimization of the newly developed devices for CE-MS coupling. Various contaminants presented in materials can cause undesirable signal suppression characteristic for electrospray ionization which was used as a source of ions for mass spectrometry and decrease sensitivity of method. Sensitivity and mechanical and chemical stability was one of the criterions for the selection of the materials used for the device construction. Finally, possible miniaturization and ability to work with small amount of samples as well as fidelity and robustness of developed instrumentation was the last important criterion aimed at during the CE-MS system design.
