The study of acute myeloid leukemia engraftment in NOD scid gamma mouse

Abstract:

Acute myeloid leukaemia belongs to haematological malignancies with a very poor prognosis. The onset of the disease is rapid, with no specific symptoms, which is probably related to the diagnosis of the disease in an advanced and difficult-to-treat stage. It is an aggressive disease that requires aggressive therapeutic procedures. Due to the heterogeneity of the disease current studies are primarily concerned with AML clonality and understanding the process of leukaemogenesis. In this area, in vivo studies using immunodeficient NSG mouse models represent great benefit and allow for the observation of AML under otherwise irreproducible conditions. The aim of this thesis was to compare phenotypes of clinical samples with phenotypes of xenotransplanted samples, and to further compare mutation profiles of clinical samples before and after xenotransplantation and to determine the overall similarity of leukaemia cells in the patient and in the mouse. Six diagnostic samples and one from relaps were analysed and successfully transplanted into mice. Our results show phenotypic and clonal similarity between leukaemia cells in the patient and in the mouse. Mutations present in the dominant clones were also present in mice after xenotransplantation. Changes occurred at the level of sub-clonal mutations. The expected benefit of the thesis is the possibility of studying the clinical response of specific patients or testing of therapy.

Abstract:

Akutní myeloidní leukémie se zařazuje mezi hematologické malignity s velmi špatnou prognózou. Nástup onemocnění je rychlý, bez specifických příznaků, s čímž pravděpodobně souvisí diagnostika onemocnění v pokročilém a těžko léčitelném stádiu. Jde o agresivní onemocnění, které vyžaduje agresivní terapeutické postupy. Protože mluvíme o velmi heterogenním onemocnění, současné studie se primárně zaobírají klonalitou AML a pochopením procesu leukemogeneze. V této oblasti jsou velkým přínosem in vivo studie pomocí imunodeficientních NSG myších modelů, které umožňují sledovat AML za jinak nenapodobitelných podmínek. Cílem diplomové práce bylo porovnat fenotypy klinických vzorků s fenotypy xenotransplantovaných vzorků, dále porovnat mutační profily klinických vzorků před a po xenotransplantaci a určit celkovou míru podobnosti leukemických buněk u pacienta a u myši. Analyzovaných bylo šest diagnostických vzorků a jeden vzorek z doby relapsu, které byly úspěšně transplantovány do myší. Naše výsledky ukazují fenotypovou i klonální shodu mezi leukemickými buňkami u pacienta a u myši. Mutace přítomné v dominantních klonech byly přítomné i v myších po xenotransplantaci. Změny nastaly na úrovni subklonálních mutací. Očekávaným přínosem práce je možnost studie klinické odpovědi konkrétních pacientů anebo testování terapie.
