Seasonal dynamics of spread of the causal agent of sooty bark disease in Brno - Sadová

Abstract:

Sazná nemoc kůry je způsobena parazitickou houbou Cryptostroma corticale (Ellis & Everhart) Gregory & Waller, která v hostiteli (nejčastěji javoru klenu) přečkává v endofytické fázi. Po vystavení hostitele stresu především suchem, se projeví její silné patogenní účinky, které mohou být pro hostitele fatální. Na výzkumné ploše v Soběšicích proběhl od jara do podzimu odběr vzorků částic ze vzduchu pomocí auto-matického volumetrického lapače spor. V těchto vzorcích byl následně pomocí extrakce DNA a real-time PCR zjištěn počet spor C. corticale. Následně byla porovnávána závis-lost výskytu spor na různých meteorologických veličinách. Za podobných podmínek bylo pozorováno více spor v jarních měsících než v podzimních.

Abstract:

Sooty bark disease is caused by a parasitic fungus Cryptostroma corticale (Ellis & Ever-hart) Gregory & Waller, which survives in its host (mostly sycamore maple) in engophy-tic stage. After exposing the host to stress, especially drought stress, its strong pathoge-nic effects prevailes, which may be fatal for the host. At the research area in Soběšice, aerobiological sampling took place from spring to autumn using automatic volumetric spore trap. DNA extraction and real-time PCR were used to determine the number of C. corticale spores in the samples. The effect of meteorological factors on the spore amounts was determined. More spores were observed in the spring months than in the autumn months under similar conditions.
