Microevolutionary processes of apomictic Rubus subgenus Rubus

Abstract:

Podrod Rubus patři k nejkomplikovanějším agamickým komplexům krytosemenných rostlin v Evropě, který v současné době zahrnuje přibližně 750 druhů. Polyploidie, hybridizace a apomixie kombinovaná se sexualitou dává vzniknout velkému množství morfologicky variabilních biotypů s různými stupni ploidie a mírou asexuálního rozmnožování. V předkládané práci jsou odkrývány evoluční procesy středoevropských ostružiníků studiem způsobu rozmnožování a genetické variability vybraných biotypů a taxonů. V první části práce jsou shrnuty doposud uváděné informace o ploidních úrovních tří sekcí podrodu Rubus (Rubus, Corylifolii a Caesii) a stanovena úroveň apomixie / sexuality a genetické variability zástupců tří sérií (Discolores, Glandulosi and Radula - sekce pravých ostružiníků Rubus), které se podílejí na evolučních procesech středoevropských ostružiníků. V rámci všech sekcí se nejčastěji vyskytují tetraploidní druhy. Evolučně původnější sekce Rubus zahrnuje také diploidní a triploidní druhy, zatímco sekce Corylifolii je častěji než ostatní skupiny zastoupena odvozenými penta- a hexaploidními druhy. Mezi studovanými ostružiníky je sexuální způsob rozmnožování plně zachován pouze u diploidního Rubus ulmifolius a převažuje rovněž u tetraploidní ser. Glandulosi, ačkoliv u zástupců této série byla detekována snížená produkce redukovaných vajíček v oblasti jihozápadního Českého masívu. Všechny ostatní studované tetraploidní druhy mají různou úroveň fakultativní apomixie. Oproti tomu triploidní ostružiníky nejsou schopné produkovat redukované gamety, ačkoliv si zachovávají schopnost oplození vaječné buňky. Následkem toho příložitostně vzniká potomstvo s vyšší ploidní úrovní. Tímto způsobem vznikly pravděpodobně některé penta- a hexaploidní druhy ze sekce Corylifolii. Při kultivačních pokusech bylo zjištěno, že rozmnožování R. bifrons (ser. Discolores) je ovlivněno vnějšími podmínkami jako je teplota nebo přítomnost cizího pylu. Vysoká heterozygotnost a nízká genetická variabilita apomiktických ostružiníků naznačuje jejich hybridní původ, kdy každý klon vznikl pouze jednou a v průběhu času akumuloval somatické mutace. Ze studia chloroplastů jednoznačně vyplývá, že série Glandulosi se chová ve všech hybridizačních procesech jako mateřská rostlina. Zbytková sexualita apomiktů má zanedbatelný vliv na evoluci středoevropských ostružiníků, ačkoliv jejich křížení se sérií Glandulosi má zřejmě vliv na způsob rozmnožování sexuálních taxonů.

Abstract:

The subgenus Rubus (also known as brambles) is one of the most complicated agamic complexes of angiosperms in Europe comprising approximately 750 species. Polyploidy, hybridization and apomixis combined with sexuality generates a high amount of morphologically different biotypes with different ploidy levels and mostly asexual reproduction. In order to clarify the evolutionary processes of central European brambles the reproduction systems and genetic variability of seleceted biotypes and taxa have been studied. We assessed the knowledge of ploidy levels of three sections (Rubus, Corylifolii and Caesii) of European brambles, evaluate the level of apomixis / sexuality and determine the genotypic variability of members of three series (Discolores, Glandulosi and Radula; all belonging to section Rubus), which are included in the evolutionary processes of central European brambles. The most common ploidy level of all three sections is tetraploid. Basic group of the subgenus is section Rubus, which also contains the evolutionary primitive diploid and triploid species. Compared to it, the section Corylifolii contains more advanced ploidy levels (5x and 6x). The sexual reproduction is maintained in the studied diploid Rubus ulmifolius and tetraploid ser. Glandulosi, although the latter shows a decrease in the production of reduced egg cells in the region of the southwestern Bohemian Massif. All of the other tetraploid taxa which were studied are facultative apomicts with different proportions of sexuality. Triploid brambles are not able to produce reduced gametes, although they retain the ability of cross-fertilization, leading to an occasional increase of ploidy levels in the offspring. The hypothetical origin of the section Corylifolii from such a hybridization event is supported by the generally higher ploidy levels of its members. We detected that reproduction of R. bifrons (ser. Discolores) is influenced by environmental conditions such as temperature and/or presence of out-crossed pollen. High heterozygosity and low genotypic variability of apomictic species suggest their single hybrid origin followed by an accumulation of scarce somatic mutations. In the hybridization processes the sexual ser. Glandulosi always acts as the mother, as indicated by the chloroplast haplotypes. Residual sexuality of apomicts seems to have negligible effect on the evolution of central European brambles, although crossing with sexuals might affect the reproduction of the sexual taxa. The driving force of evolution and speciation of subgen. Rubus in central Europe is the sexuality of ser. Glandulosi.
