Enzymatic determination of methanol in the presence of ethanol

Abstract:

Bachelor thesis describes basic attributes of methanol, its toxicity, legislative definition of alcoholic beverages and methods of determinating methanol in alcoholic beverages. Thesis describes most used methods of its determination and the new enzymathic methods as well. Special attention is dedicated to catalase, its structure and mechanism of its effects. Attention is paid to methanol and multiple methods of its determination, including oxidation of methanol to formaldehyde by catalase. Formaldehyde is then incorporated into the molecule of dihydropyridine derivative, using Hantzsch reaction. Experimental part describes spectrometric measurments of Hantzsch reactions using multiple diketones and compares its reactivities with formaldehyde and acetaldehyde. High selectivity of acetoacetanilide and 4-chloroacetoacetanilide to formaldehyde was confirmed. On the contrary, acetylacetone was confirmed as more selective to acetaldehyde.

Abstract:

Bakalářská práce se zabývá methanolem, jeho toxickými účinky, legislativní definicí alkoholických nápojú a metodami stanovení methanolu v nich. Souhrně popisuje i nové enzymatické metody a podrobné vlastnosti použitého enzymu, katalasy. Pozornost je věnována struktuře katalasy a mechanizmu jejího účinku. V teoretické části jsou uvedené vlastnosti methanolu a rozmanité metody jeho stanovení, včetně experimentálně testované přemeny pomocí katalasy na formaldehyd a jeho následné inkorporace do derivátu dihydropyridínu pomocí Hantzschovy reakce. V experimentální části byli provedeny spektrofotometrické meření Hantzschovych reakcií s různými diketony a porovnány ich reaktivity s oxidačnými produkty methanolu a ethanolu, resp. formaldehydem a acetaldehydem. Potvrzená byla vysoká selektivita acetoacetanilidu a jeho chlorovaného derivátu k formaldehydu v Hantzschovi reakci, na rozdíl od acetylacetonu, který se projevil vyšší selektivitou ku acetaldehydu.
