Preparation and characterization of yeast strains defective in RNA processing and degradation

Abstract:

Proteiny Trf4 a Trf5 jsou součástí multiproteinového polydenylačního komplexu TRAMP, který je spolu s exosomem zodpovědný za kontrolu kvality RNA v eukaryotických buňkách. Úkolem Trf4p a Trf5p je beztemplátová polyadenylace aberantních či přebytečných molekul RNA, které jsou následně rozpoznány a degradovány ve vlastním rozkladném centru – exosomu. Další studie prokazují jejich roli při mitotickém dělení a replikaci DNA. Dosud však schází spolehlivý systém, který by pomohl objasnit mechanismy rozpoznávání cílových molekul a blíže specifikovat fyziologickou funkci Trf4p a Trf5p. Tato bakalářská práce se zabývá problémem heterologních expresních systémů S. cerevisiae s možností regulovatelné exprese genu TRF4. Jedná se zejména o vektory obsahující tetracyklinové, galaktosové a methioninové promotory. Cílem práce bylo zkonstruovat takový expresní vektor, který by dovoloval striktně řízenou expresi studovaného genu, aniž by hlouběji zasahoval do přirozeného metabolismu kvasinkové buňky. K tomuto účelu byly vytvořeny kmeny S. cerevisiae, ve kterých byl gen TRF4 pod kontrolou MET3 promotoru. Dále byla úspěšně provedena expresní analýza a test represe, které prokázaly regulovatelnost exprese studovaného genu.

Abstract:

Trf4p and Trf5p are part of multiprotein polyadenylation complex TRAMP, that is, in association with exosome, responsible for RNA quality control in eukaryotic cells. The task of Trf4p and Trf5p is non-template polyadenylation of aberrant or unnecessary RNA molecules, which are then recognized and degraded in decomposition centre – exosome. Further studies show that they are involved in mitotic division and DNA replication. However, a reliable system, which would help to clarify the mechanisms of targeting RNA substrates and thus specify physiologic function of Trf4p and Trf5p, is still lacking. The bachelor thesis focuses on the problem of heterologous expression systems of S. cerevisiae with regulatable expression of TRF4 gene. These include especially vectors with tetracyclin, galactose and methionin driven promoters. The aim of this work was to construct such an expression vector, which would allow a strictly regulated expression of target gene, without further interference in the natural metabolism of the yeast cell. Strains of S. cerevisiae, where the TRF4 gene is under the control of MET3 promoter, have been constructed for this purpose. Moreover, the expression analyse and the test of repression, were successfully performed. This analyses prooved the adjustability of the expression of the target gene.
