Abstract:

Propojení elektrochemických metod s biochemických a biologickými vědmíni disciplínami může pomoci vyřešit mnoho výzev v obou odvětvích. Interpretace výsledků elektrochemických analýz může pomoci lépe pochopit různé biochemické a biologické procesy. V současnosti jsou v elektrochemii používány biochemické a biologické komponenty jako biorekogniční elementy biosenzorů. Z tohoto důvodu je velice důležité rozumět dějům, které probíhají na rozhraní elektrody a biologické nebo biochemické složky. Například enzymatické biosenzory jsou schopny selektivní a vysoce citlivé kvantifikace biomolekul jakými jsou například glukóza nebo peroxid vodíku. Enzymy se mohou potýkat s problémy se stabilitou, která může být negativně ovlivněna změnou pH nebo teploty. Z tohoto důvodu je snaha nahradit enzymy syntetickými sloučeninami, které vykazují lepší stabilitu. Dobrý potenciál pro substituci přírodních enzymů vykazují nanočástice kovů a komplexy kovů. Dalším vhodným nedestruktivním nástrojem pro studium biologických procesů je Skenovací elektronová mikroskopie (SECM), která může být využita pro studium biologických interakcí in situ. Prozatím byla pomocí SECM sledována životaschopnost buněk, které byly vystaveny vlivu různých chemických činidel. Pro studium biologických a biochemických procesů na rozhraní elektrody a buněk byl použit SECM pracující v amperometrickém nebo potenciometrickém módu.

Abstract:

Incorporation of electrochemistry to biochemistry and biology can respond to many challenges. In fact, the electrochemical signals can be applied to get insight to biological and biochemical processes. On the other hand, the biochemical and biological compounds have been used in electrochemistry, particularly in biosensing and bioassays. Therefore, it is highly demanded to investigate more in the interfaces of electrochemistry with biology and biochemistry. As an instance, biosensing based on the enzymes are already known as the sensing platform with high selectivity and sensitivity used for quantification of various biomolecules such as hydrogen peroxide and glucose. However, the recent researchers try to substitute the enzyme with a chemically synthesized compound to overcome the problems due to instability of natural enzyme in severe conditions of temperature and pH. With this regard the metal complexes and the nanomaterials were shown to be potential alternatives to replace the natural enzymes. Moreover, electrochemistry offers a promising tool to study the biological processes, where scanning electrochemical microscopy (SECM) can be utilized as a non-destructive tool for in situ analysis of the biological interactions. Indeed, SECM have been successfully used to track the cell viability affected by different chemical interactions and to observe the interfacial reactions which may alter the cell viability. Hence, the application of SECM either in potentiometric or amperometric mode was employed in study the biochemical and biological processes.
