Generation and characterization of polyclonal antibodies against parts of positive elongation factors

Abstract:

V této diplomové práci se věnuji testování polyklonálních protilátek z králičích sér (CDK13-C, CDK13-N, CycK-1-61) a kozího séra (CDK13-532) pro metody Western blot, imunoprecipitace, nepřímé imunofluorescence a imunohistochemie. Tato polyklonální séra byla připravena proti cyklinu K a kináze CDK13. Důvodem byl fakt, že v období zadání diplomové práce, nebyly komerčně dostupné protilátky proti cycklinu K a CDK13. Proteiny cyklin K a CDK13 obecně hrájí klíčovou roli v regulaci elongační fáze eukaryotické transkripce, která se zdá být zcela zásadním kontrolním bodem v procesu eukaryotické transkripce. Vlastní charakterizace sér byla provedena jak na lidských buňkách HCT116, U2OS tak i myších kmenových buňkách R1. Ukázala jsem, že séra pro CDK13 se dají využít k detekci proteinové hladiny a k precipitaci endogenní CDK13 v lidských a myších buňkách. Dále jsme prokázala, že sérum CycK-1-61 není vhodné pro detekci proteinové hladiny cyklinu K v lidských a myších buňkách, ale dá se použít k precipitaci cyklinu K v obou typech buněk. Bohužel se ukázalo, že ani jedno z připravených sér není vhodné pro metodu imunocytochemie nebo imunohistochemie. Následně byla provedena izolace frakce IgG protilátek z kozího séra CDK13-532 a byl proveden test, zda přečištění vedlo ke snížení nespecifických signálů v imunoblotu. Na základě mnou provedených experimentů byla séra vyvinuta k detekci CDK13 a cyklinu K a jsou nadále využívána v laboratoři školitele pro účely imunoblotu a imunoprecipitace.

Abstract:

This thesis is focused on testing the polyclonal antibodies from rabbit sera (C-CDK13, CDK13-N-CycK 1-61) and goat serum (CDK13-532) in use of Western blot, immunoprecipitation, indirect immunofluorescence and immunohistochemistry. The polyclonal sera were prepared against cyclin K and kinase CDK13. The reason was the fact that at the beginning the diploma work, there were not commercially available antibodies against cycklin K and CDK13. Proteins cyclin K and CDK13 generally play a key role in the regulation of eukaryotic transcription elongation phase, which seems to be quite a major control point in the process of eukaryotic transcription. Own characterization of sera was performed on both human HCT116 cells, U2OS and mouse stem cells R1. I showed that the serum for CDK13 can be used to detect protein levels and precipitation of endogenous CDK13 in human and murine cells. Furthermore, we demonstrated that serum CycK-1-61 is suitable for the detection of protein levels of cyclin K in human and murine cells, but can also be used to precipitate cyclin A in both cell types. None of the prepared sera are suitable for immunocytochemistry or immunohistochemistry. Subsequently, fractions of IgG were isolated from goat serum CDK13-532 and were tested whether purification led to a reduction of non-specific signals in immunoblot. The sera were developed for detection of CDK13 and cyclin K and are used in the lab of my supervisor for immunoblotting and immunoprecipitation.
