Influence of nitrate supplementation on oxygen uptake during exercise.

Abstract:

V této práci byl zkoumán vliv suplementace nitrátů na spotřebu kyslíku při svalové práce. Ve dvojitě zaslepené, zkřížené studii deset zdravých mužů (věk 26,9 ± 5,72, výška 185 ± 5,23, váha 81,75 ± 5,66) podstoupilo 8 laboratorních vyšetření v průběhu 4 – 5 týdnů. Během první návštěvy absolvovali rampový zátěžový test na bicyklovém ergometru pro určení respiračního prahu a maximální aerobní kapacity. Následovalo náhodné rozdělení do dvou skupin, kde dostali šestidenní suplementaci džusu z červené řepy nebo placeba. 4., 5. a 6. den suplementačního období absolvovali zátěžový test na stejném přístroji pro určení dynamiky VO2 při různých intenzitách zátěže. Přestávka mezi suplementačním obdobím byla 12 dní. Hodnoty výsledků po podání účinné látky nebo placeba byly porovnány neparametrickým Wilcoxonovým párovým testem, kdy hladina významnosti byla stanovena p< 0,05. Při podání účinné látky se zvýšila hladina plazmatických nitrátů, metabolická přeměna na nitrity a dále na NO však neproběhla. Hodnoty sledovaných parametrů: spotřeba kyslíku, tepový kyslík, poměr respirační výměny, srdeční frekvence, krevní tlak a zatížení cirkulace byly ve všech případech zvýšené při podání účinné látky oproti placebu. Pozitivní vliv suplementace nitráty na ekonomiku spotřeby kyslíku při svalové práci tak nebyl prokázán.

Abstract:

In this diploma thesis I was examine the influence of nitrate supplementation on oxygen uptake during the exercise. In a double-blind crossover study, the ten health men (age 6,9 ± 5,72, height 185 ± 5,23, weight 81,75 ± 5,66) take the 8 laboratory tests over four to five week period. During the first appointment the subjects were perform a ramp incremental exercise test for determination of the peak VO2 and respiratory threshold. Then the subjects were randomly assigned in a crossover design where they receive 6 days of dietary supplementation with either beetroot juice or placebo. On days 4, 5 and 6 of the supplementation periods, the subjects were completed step exercise tests on the same machine for the determination of pulmonary VO2 dynamics in different intensity of endurance. The break between the supplementation periods was 12 days. The values of the results after nitrate supplementation or placebo were compare with non-parametric Wilcoxon pair test, when the level of consequence was specify at p<0,05. After nitrate supplementation the level of plasma nitrates was higher, but the transformation at nitrites and also at NO did not happen. The values of measured parameters: oxygen uptake, pulsation oxygen, respiratory exchange ratio, heart rate, blood pressure and the loading of circulation were higher in all cases when the subjects gets nitrate supplementation compared to placebo. The positive influence of nitrate supplementation on economy of oxygen uptake during the exercise was not approve.
