Preparation of metallodrugs with steroid ligands

Abstract:

Od doby co se ke klinickým testům dostaly dva nejznámější komplexy Ru(III), NAMI a NAMI-A, už uplynulo 12 roků. Během těchto let se mnoho výzkumných skupin po celém světě zabíralo vývojem analogických komplexů s potencionální biologickou aktivitou. V této práci přinášíme spojení dvou perspektivních látek - komplexů Ru(III) s derivatizovanými steroidními ligandy. V teoretické části práce jsou shrnuty základní znalosti o metaloléčivech a steroidech. Samostatná kapitola je věnována popisu vzniku inkluzních komplexů steroidů s cyklodextriny a jejich aplikací ve farmacii. V experimentální části práce je popsaná příprava prekurzorů Ru(III) komplexů s organickým, sodným a draselným protiiontem. Z těchto prekurzorů byly dále připraveny komplexy s deriváty žlučových kyselin a to a) typu NAMI-A, tedy s ligandem jako protiiontem, kde bylo připraveno 6 nových látek, b) typu NAMI, tedy se sodným protiiontem, kde byly připraveny 3 látky. V případě vybraných komplexů typu NAMI (Na) jsme se dále zabývali jejich samotným zavedením do cyklodextrinových kavit. Komplexy hostitel-host byly studovány titračními experimenty pomocí 1H NMR spektroskopie, předběžné výsledky těchto studií jsou uvedeny v závěru práce.

Abstract:

Twelve years passed since two most important Ru(III) complexes, NAMI and NAMI-A, entered clinical trials, in the meantime, many research groups all around the world were dealing with development of analogous complexes with potential biological activity. In this work, we are introducing a link between two prospective compounds – complexes of Ru(III) and steroid ligands. In the theoretical part, we present basic facts about metallodrugs and steroids. Special chapter is dedicated to description of formation of inclusion complexes of steroids with cyclodextrins and their application in pharmacy. In the experimental part the preparation of precursors of Ru(III) complexes with organic, sodium, and potassium counterion is described. From these precursors a) NAMI-A type complexes with ligand as counterion (6 compounds), b) NAMI type complexes with sodium counterion (6 compounds) and potassium counterion (3 compounds) were prepared. With selected NAMI (Na) type complexes their host-guest chemistry with cyclodextrins was studied using 1H NMR titration experiments, some preliminary results are listed in the latter part of this work.
