Role of the p62 protein in exosomal secretion related to the level of autophagy

Abstract:

Autofagie je proces recyklace makromolekul a organel, který má klíčovou úlohu v udržování buněčné homeostázy. Jedná se o vysoce konzervovaný proces, přísně regulovaný produkty autofagických genů. Jako významný autofagický adaptor slouží protein p62/SQSTM1, který označuje a transportuje substrát určený k degradaci selektivní autofagií. Tento protein se v důsledku inhibice autofagie hromadí a při její indukci odbourává. Z tohoto důvodu může být využit jako marker autofagické aktivity v buňkách. Autofagie sdílí některé regulační komponenty s mechanismem exozomální biogeneze a sekrece, jedná se tedy o dva úzce spjaté procesy. Cílem této práce bylo porovnat míru autofagie a hladiny proteinu p62 za různých stresových podmínek s mírou exozomální sekrece sledované pomocí hladin tetraspaninů u buněčné linie spinocelulárního karcinomu. Výsledkem je stanovení vlivu autofagických induktorů a inhibitorů na sekreci exozomů, která je klíčová pro komunikaci nádorových buněk.

Abstract:

Autophagy is defined as a process that recycles macromolecules and organelles and plays a pivotal role in the maintenance of homeostasis. This process is highly conserved and strictly regulated by products of autophagy genes. Protein p62/SQSTM1 acts as an important autophagy adaptor, which marks and transport autophagic cargo to be degraded by selective autophagy. Protein p62 is accumulated during autophagy inhibition and is degraded when autophagy is induced. Therefore, this protein is used as an indicator of the level of autophagy in cells. The process of autophagy is closely related to exosomal biogenesis and secretion, as they both share certain regulatory components. This thesis aims are to compare the level of autophagy and p62 expression by the cell line of spinocellular carcinoma under different stress conditions to the expression of tetraspanins, which are exosomal markers. As the result of this thesis, we determine the influence of autophagy inducers and inhibitors on exosomal secretion, a key mechanism for cellular communication in cancer cells.
