Examination methods of gene variability in fish – comparison of SSCP and HRM

Abstract:

Genes of the major histocompatibility complex (MHC) encode surface glycoproteins that are capable of binding antigenic peptides and subsequently present them to T-lymphocytes, which they contribute of causing the immune response. They are highly polymorphic, and we find them in all jawed vertebrates. It is now clear that MHC is not just transplantation factor but has a crucial role in resistance to pathogens and infectious diseases (Rakus et al., 2009b). The presented thesis provides a brief overview of existing knowledge about MHC glycoproteins and their genes based on the studied literature. It further describes the appropriate methods to study the structural variability of MHC genes in fish, particularly focusing on SSCP and HRM. In the experimental part of the thesis there were samples of DAB genes of species Parachondrostoma toxostoma and Chondrostoma nasus analyzed using both methods and the obtained results were compared.

Geny hlavního histokompatibilného komplexu (MHC) kódují povrchové glykoproteiny, které jsou schopné vázat antigenní peptidy a následně jich prezentovat T-lymfocytům, čím se podílejí na vzniku imunitní odpovědi. Jsou vysoko polymorfní a najdeme je u všech čelistnatých obratlovců. Dnes je už jasné, že MHC je nejen transplantačním faktorem ale má i rozhodující úkol v rezistenci vůči patogenům a infekčním onemocněním (Rakus et al., 2009b). Předložená bakalářská práce podává na základě nastudované literatury stručný přehled dosavadních poznatků o MHC glykoproteinech a jejich genech. Dále popisuje vhodné metody ke studiu strukturní variability MHC genů u ryb, především se zaměřuje na SSCP a HRM. V experimentální části bakalářské práce byly analyzovány vzorky DAB genů druhu Parachondrostoma toxostoma a Chondrostoma nasus pomocí obou metod a získané výsledky byly porovnané.
