The effect of platinum drugs to healthy and cancer tissue; insights in the alteration of the amino acid metabolism

Vliv platinových léků na zdravé a rakovinné tkáně; pohled na proměny metabolismu aminokyselin

Abstract:

Aminokyseliny jsou organické sloučeniny plnící četné funkce, a zatímco se u zdravých buněk a tkání udržuje jistá rovnováha těchto funkcí, rakovinné buňky mají tendenci využívat aminokyseliny k podpoře jejich šíření a tudíž i růstu nádoru. V této práci představujeme vliv léků na bázi platiny, cisplatiny a platinových nanočástic, na zdravé a rakovinné buňky a tkáně, a rovněž odhalujeme narušení metabolismu aminokyselin v důsledku léčby. Cisplatina zvětšuje obsah aminokyselin ve zdravých i rakovinných buňkách, dopad platinových nanočástic na aminokyseliny ve zdravých buňkách je sice zanedbatelný, v rakovinných buňkách je však vyčerpávající. U prošetřených tkání, jako jsou játra, ledviny a mozek, jsme zaregistrovali jisté vyčerpání aminokyselin, které je nejzřetelnější u jater, tento vliv je však méně viditelný u rakovinné tkáně. Dále léčba způsobila přerušení cyklu třikarboxylových kyselin u rakovinných buněk, a přitom založila jinou půdu pro cisplatinu a platinové nanočástice.

Abstract:

Amino acids are organic compounds with numerous functions, and while a certain balance of these functions is sustained in healthy cells and tissues, cancer cells tend to adapt the utilization of amino acids for the processes needed to support their proliferation and tumor growth. Herein, we present the effect of platinum-based drugs, cisplatin and platinum nanoparticles in both healthy and cancer cells and tissues, and we reveal the disruptions of the amino acid metabolism by the administered treatments. Cisplatin increases the contents of the amino acids in healthy and in cancer cells, whereas the platinum nanoparticles have negligible impact on the amino acid in healthy cells but present a depleting effect of amino acids in cancer cells. The investigated tissues, liver, kidney, and brain, also record a certain depletion of amino acids, which is mostly apparent in liver, however, the effects are far less obvious than in tumor tissue. In addition, the treatments caused disruptions of the tricarboxylic acid cycle in cancer cells, displaying a different landscape of targets for cisplatin and platinum nanoparticles.
