The protein inhibitors of peptidases from Eudiplozoon nipponicum and their preparation in recombinant form by using the cell-free expression systems

Abstract:

Tato práce je v návaznosti na bakalářskou práci zaměřena na funkční proteiny parazitických červů – helmintů. Konkrétně jsme se zaměřili na modelový organismus Eudiplozoon nipponicum, který je řazen do třídy Monogenea. Tato skupina parazitů nebyla dosud z hlediska jejich biochemie prakticky zkoumána. V rámci experimentální části této práce jsme se proto zabývali genovou expresí funkčních proteinů E. nipponicum, konkrétně inhibitory serinových peptidáz – serpiny, které hrají roli v mnoha fyziologických procesech (srážení krve, fibrinolýza, vznik zánětu apod.). Pro samotnou expresi byly z transkriptomických dat selektovány sekvence dvou domnělých serpinů, které jsme se pokusili připravit v rekombinantní podobě. Pro tyto účely byly zvoleny dva přístupy – exprese pomocí klasického buněčného systému, založeného na bakteriích E. coli a tzv. nebuněčná (cell-free) in vitro exprese, založená na lyzátu z E. coli. Měli jsme tak možnost v rámci experimentu tyto postupy vzájemně porovnat.

Abstract:

This thesis is focused on functional proteins of parasitic worms – helminths. As an experimental model organism we used Eudiplozoon nipponicum, belonging into class Monogenea. From biochemical point of view, this groupe of parasites has not been investigated yet. Therefore we decided to express the E. nipponicum selected genes of functional proteins, serine peptidases – serpins. These molecules play a role in wide range of physiological processes (blood clotting, fibrinolysis, inflammation etc.). The sequences of two presumed serpins were selected from the transcriptomic data and then we started to prepare them in the recombinant form. We chose two expression systems, classical bacterial expression system, based on Escherichia coli and cell-free in vitro expression system, based on E. coli lysate. The advantages and disadvantages of both systems were evaluated.
